IL-1β对体外培养MRC-5细胞增殖、表型转化的影响

摘要 目的探讨外源性白介素-1β(IL-1β)对体外培养的人胚肺成纤维细胞(MRC-5)表型转化的影响。方法体外培养人胚肺成纤维细胞(MRC-5),以不同浓度IL-1β(0、0.1、1、5、10ng/ml)刺激48h和同一浓度(10ng/ml)IL-1β刺激不同时间(0、24、48、72h);采用CCK-8法检测细胞增殖情况,半定量反转录聚合酶链式反应法(RT-PCR)、Western blot检测细胞表型转化标志物α-平滑肌肌动蛋白(α-SMA)的mRNA和蛋白水平。结果 IL-1β以浓度和时间依赖性方式调节MRC-5细胞α-SMA mRNA和蛋白表达。结论 IL-1β可能通过促进肺成纤维细胞增殖和肺成纤维细胞-肌成纤维细胞的细胞表型转化,促进气道炎症后的重塑进程。 Objective To observe the effect of cell proliferation and phenotypic differentiation on MRC - 5 stimulated with IL - 1β in vitro. Methods MRC - 5 were cultured in vitro after stimulation either with different concentrations (0,0.1,1,5,10ng/ml) of IL - 1for 48h, or with the same concentration (10ng/ml) for different time (0, 24, 48, 72h). Then cell proliferation, the expression of α - SMA -mRNA and protein were detected by CCK -8 cell proliferation assay, semi -quantitative reverse transcription polymerase chain re- action (RT - PCR) and Western blot. Results IL - 1β induced cell proliferation, the gene expression and protein production of α - SMA on MRC - 5 in vitro. Conclusion Lung fibroblasts may play an important role in the process of airway remodeling after inflammao- tion, by the means of upgrading cell proliferation and phenotypic differentiation after simulation of IL - 1β.
