摘要
目的检测MAGE基因家族的三个成员MAGE-1、MAGE-2、MAGE-3 mRNA在非小细胞肺癌(NSCLC)患者中的表达情况,探讨以该基因家族成员作为靶点对NSCLC患者进行免疫治疗的可能性。方法采用逆转录-聚合酶链式反应(RT-PCR)和DNA印迹(Southern blot)的方法,对MAGE基因家族的三个成员MAGE-1、MAGE-2、MAGE-3 mRNA在56例NSCLC中的表达情况进行研究。结果 56例NSCLC肿瘤组织中,33例表达MAGE基因,正常肺组织均不表达;MAGE-1、MAGE-2、MAGE-3 mRNA的表达例数分别为27、18、30。结论 MAGE基因家族成员在NSCLC患者中呈高频率表达,提示该基因是对NSCLC患者进行免疫治疗的适宜靶点。
Objective To determine the expression of 3 members(MAGE-1, MAGE-2, MAGE-3) of MAGE gene family in non-small cell lung cancer (NSCLC) and to explore the possibility of using these genes as targets for immunotherapy for NSCLC patients. Methods The expression of MAGE-1, MAGE-2 and MAGE-3 mRNA in 56 NSCLC samples were examined by RT-PCR and Southern blot. Results Of the 56 NSCLC samples, 33 expressed MAGE gene, whereas none of the normal lung samples expressed MAGE gene. The number of the NSCLC cases who expressed MAGE-1, MAGE-2 and MAGE-3 mRNA was 27, 18 and 30 respectively. Conclusion Members (MAGE-1, MAGE-2, MAGE-3) of MAGE gene family are expressed at a high frequency in NSCLC samples, and these genes are the suitable targets in immunotherapy for NSCLC patients.
