摘要
目的研究远志中寡糖酯类化合物的神经保护作用和探讨化合物的初步构效关系。方法采用MTT法和BrdU标记法评价寡糖酯单体化合物对神经细胞的保护作用。结果在体外皮质酮诱导的SH-SY5Y神经细胞损伤模型上,化合物tenifoliside A、tenuifoliose J、tenuifolioses A、1-o-(E)-Benzoyl-[3-o-(E)-Alphatolluyl]-β-D-fructofuranosy-(2→1)-[β-D-glu-copyranosyl-(1→2)]-α-D-gluc-opyranosid表现出了很好的神经保护作用,并呈一定的剂量依赖性。结论远志寡糖酯类化合物对皮质酮诱导损伤的SH-SY5Y神经细胞有保护作用,其保护作用强弱与化合物类型及其浓度,化合物的母核结构,取代基位置和取代基种类有关。
Objective To study nerve protection by sucrose esters of Polygala tenuifolia Wild. and discuss the preliminary structure-activity relationship. Methods The nerve protection property of sucrose esters was evaluated using MTT assay and BrdU mark method. Results Tenifoliside A, tenuifo- liose J, tenuifolioses A, 1-o- (E) -Ben- zoyl- [ 3-o- ( E ) -Alphatolluyl ] -β -D-fructofu- ranosy- ( 2→1 ) - β -D-glucopyranosyl - ( 1→2 ) ] - a -D-glucopyranosid showed dose-dependent protection activites on SH-SYSY cells from the lesion done by corticosterone. Couclusion Some of the sucrose esters from Polygala tenuifolia Wild. can protect SH-SYSY nerve cells exposed to corticosterone. The amount of protection depends on the compound type, concentration, structrure of the nucleus and location of substituents.
