摘要
本文报道了海滨锦葵块根中皂苷及多糖的分离、纯化方法,以及皂苷的体外抑制肿瘤细胞增殖活性、多糖的免疫活性的检测。采用超声波法对块根中的皂苷和多糖进行粗提;通过大孔树脂柱层析法对得到的皂苷粗提取物进行纯化;皂苷经酸解得到皂苷元部分;采用DEAE-纤维素柱层析对粗多糖溶液进行提纯。利用MTT比色法测定皂苷对肿瘤细胞HepS增殖的影响以及多糖对胸腺淋巴细胞、脾淋巴细胞增殖的影响。结果表明,海滨锦葵块根皂苷提取率为0.089%,粗多糖提取率为5.10%;海滨锦葵块根皂苷及皂苷元有抑制HepS增殖的作用,皂苷元效果更好;海滨锦葵块根多糖能显著促进淋巴细胞体外增殖,增强体液免疫力。
This paper reported the extraction,separation and purification of saponin and polysaeeharides from root tuber of Kosteletzkya virginica. Also, the antiproliferative effect of saponin and the immune-modulating action of polysaecharides were tested. We used ultrasound to extract crude saponin and polysaceharides, and then we use maeroporous resin to pu- rify saponin. Saponin was broke down to get the genipin by sulfuric acid. Polysaccharides were purified by column chro- matography on DEAE-cellulose. The effects of saponin on the proliferation of mouse hepatoma cell (HepS) and effects of polysacchafides on the proliferation of thymus lymphocyte and spleen lymphocyte were detected by MTY colorimetric as- say. The results showed that, the saponin extraction rate was 0. 089% and the polysaocharides extraction rate was 5. 10%. Saponin and genipin can inhibit the proliferation of HepS significantly, however genipin got better effect. Polysac- charides can promote the proliferation of thymus lymphocyte and spleen lymphocyte, which shows good immune-enhan- cing action.
