摘要
目的:观察鞘内注射高浓度医用臭氧对大鼠运动功能和脊髓组织中Bax、Bcl-2基因表达的影响。方法:SD大鼠16只,随机均分为2组(n=8),分别建立大鼠鞘内置管模型。对照组(C组)仅行鞘内穿刺置管不注射;医用臭氧组(O3-60组):鞘内置管后经导管注射60μg/mL医用臭氧10μL。麻醉前及注射医用臭氧后24 h分别对两组大鼠进行BBB运动功能评分;然后立即断颈取材,采用Western blot测定腰膨大处脊髓组织Bax、Bcl-2蛋白表达水平。结果:与对照组相比,鞘内注射高浓度医用臭氧24 h后,臭氧组大鼠后肢BBB运动功能评分(Basso Beattie andBresnahan locomotor rating scales,BBB scores)显著降低(P<0.05);脊髓Bax蛋白表达显著增加(P<0.05),Bcl-2/Bax比值显著降低(P<0.05)。结论:高浓度医用臭氧鞘内注射可损伤大鼠运动功能,其损伤可能与神经细胞凋亡有关。
Objective: To investigate the effects of intrathecal administration of high concentration of ozone on the animal behavior and protein expression of Bax and Bcl-2 in spinal cord of rats. Methods: Sixteen healthy adult Sprague-Dawley rats were randomly divided into two groups (n = 8): the control group (Group C) and the 60μg/mL 03 group (Group O3-60i. In Group C, rats were intrathecally catheterized without any medication, while in the Group 03-60, 60 μg/ml oxygen-ozone 10 μL was administered intrathecally. The BBB locomotor rating scales (the BBB score) were recorded before anesthesia and 24h after 03-60 administered. The lumbar spinal cord was collected to detect the protein expression of Bax and Bcl-2 by western blot. Results: After intrathecal 03-60 administration, compared with Group C, the BBB score of Group 03-60 was significantly lower (P 〈 0.05). The expression of Bax was significantly higher (P 〈 0.05), and the ratio of Bax/Bcl-2 of Group 03-60 was significantly lower (P 〈 0.05). Conclusion: The intrathecal administration of high concentration of ozone might damage the behavior of rats, and the neurotoxicity of ozone may be related to the procession of neuron apoptosis.
