中药益气活血养脑法治疗老年性痴呆的基础研究 被引量:12

摘要 目的:建立拟老年痴呆(AD)动物模型,观察中药益气活血养脑法复方胶囊对AD大鼠的认知行为及海马神经元凋亡的影响。方法:选用纯系健康Wistar雄性大鼠144只,随机分为中药保护组、中药治疗组、西药对照组、模型组、假损伤组、空白组。每组24只,采用D—半乳糖致亚急性衰老与喹啉酸损害联合所致AD大鼠模型,之后进行复方胶囊治疗。检测AD大鼠跳台实验成绩及脑内超氧化物歧化酶(SOD)含量、海马神经元凋亡情况。结果:所得数据经统计学处理表明,中药益气活血养脑法复方胶囊能明显缩短AD大鼠跳台实验潜伏减少跳台电击次数,提高超氧化物歧化酶(SOD)活力,减少海马神经元损伤。结论:中药益气活血养脑法能改善痴呆大鼠记忆,提高SOD活力,减少海马神经元损伤,促进AD大鼠的智能恢复。 Objective:To establish an animal model of Alzheimer Disease in aging rat and study the effect of the method of benefiting vital energy-promoting blood flow-providing for brain of traditional Chinese medicine on Jumping stand Test,the neural apoptosis in hippocampal region and the SOD content of AD Model Rats.Methods:Select 144 pure Wistar aging rats which were divided into model group,treatment group of Chinese crude drug,protecting group of Chinese crude drug,medicine group,negative injury group and vacant group.In the experiment we adopted D-galactose and quinaldinic acid to duplicate model AD and carried on Chinese crude drug treatment on AD rats.Then we examine the biochemistry changes and the content of SOD in brains.Results:Modelling led to the reduction of model animal's learning and memory ability.After the Chinese crude drug treatment,the statistic data showed this method not only could decrease stroke times and time of Jumping stand rats and the content of SOD,but also significantly relieve the injury of the hippocamp.Conclusion:the method of benefiting vital energy-promoting blood flow-providing for brain of traditional Chinese medicine could increase the learning and memory ability of model animal,promote the restoring function of injured neuron in hippocampal region and improve the metabolism of SOD.
