摘要
目的:探讨玻璃化冷冻对人类卵巢组织卵泡形态、超微结构和组织增殖活性的影响。方法:采用12例人卵巢组织切成薄片随机分配到新鲜卵巢组(A组)和玻璃化冷冻组(B组),两组行组织学光镜和透射电镜检查卵泡形态及结构变化,免疫组化测定卵巢组织中增殖细胞核抗原(PCNA)的表达情况。结果:A、B两组中原始卵泡和初级卵泡分布比例分别为86.4%、13.6%和84.5%%、15.5%,两组比较差异无统计学意义(P>0.05)。光镜下A、B两组中形态正常的原始卵泡分别为75.3%和69.3%,两组比较差异亦无统计学意义(P>0.05);但B组冻融后初级卵泡形态异常的比例(56%)高于A组(28.2%),两组比较差异有统计学意义(P<0.05);A、B组中形态正常的原始卵泡超微结构无明显改变,B组中初级卵泡超微结构存在一定程度的改变。A、B两组卵巢中PCNA阳性表达比较差异无统计学意义(P>0.05),PCNA阳性表达主要见于原始和初级卵泡的卵母细胞、颗粒细胞和卵巢组织间质细胞。结论:玻璃化冷冻是一种较适宜保存人类卵巢组织的方法。
Objective: To evaluate the morphology and proliferation of follicles from cryopreserved human ovarian tissue by vitrification. Methods: Ovarian biopsy specimens were taken from 12 patients. The specimens were randomly distributed into fresh group (Group A) and vitrificated group (Group B). Histological examination and uhrastructural observation were performed after cryopreservation. Both were embedded in paraffin block and proliferating cell nuclear antigen (PCNA) was detected by immunohistochemical staining. Results: The proportions of primordial and primary follicles from Group A and Group B were 86.4%, 13.6% and 84.5%, 15.5%,respectively (P 〉0.05). There was no significant difference in proportions of morphologically normal primordial follicles between Group A and Group B (P 〉 0. 05 ) ; but the proportion of morphologically abnormal primary follicles was significantly higher in Group B than that in Group A (P 〈 0. 05 ). The ultrastructural studies showed that in histologically normal primordial follicles, there was no difference between Group A and Group B, while there were a few abnormalities of primary follicles in Group B. Granulsa ceils and oocytes of primordial and primary follicles and stromal cells were positive for PCNA staining both in fresh and cryopreserved ovarian tissues; there were no differences between two groups. Conclusion: Vitrification is a favorable method in human ovarian cryopreservation.
