人脑血管母细胞瘤中VEGF和bFGF的表达及与血管生成的关系 被引量:6

摘要 目的探讨血管内皮细胞生长因子(vascular endothelial growth factor,VEGF)和碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)在人脑血管母细胞瘤中的表达及与肿瘤血管形成的关系。方法应用免疫组织化学S-P法检测32例人脑血管母细胞瘤及10例正常脑组织中VEGF和bFGF表达,以CD34单克隆抗体标记瘤组织血管内皮细胞并测定微血管密度(microvessel density,MVD)。结果人脑血管母细胞瘤组织中VEGF和bFGF的表达显著高于正常脑组织(P<0.01),肿瘤组中MVD值(168.7±56.3)高于正常对照组(24.6±13.4),VEGF与bFGF在人脑血管母细胞瘤组织中表达呈正相关(r=0.541,P<0.01)。与囊性和实性血管母细胞瘤中VEGF、bFGF和MVD的差异均无统计学意义(P>0.05)。结论 VEGF和bFGF在作用机制上可能有密切的联系或存在因果关系,检测VEGF和bFGF可以为脑血管母细胞瘤的发病机制研究及临床治疗提供新的思路。抗VEGF及抗bFGF联合治疗可能成为治疗脑血管母细胞瘤的新方法。 Objective To investigate the expression and clinical significance of vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) in hemangioblastoma and the relationship with angiogenesis in tumor. Methods Streptavidin-Peroxidase ( S-P ) immunohistoehemical technique was applied to examine and analyse the expression of VEGF and bFGF in 32 cases of hemangioblastoma and 10 cases of normal brain tissue. The endothelial cells of blood vessels within the tumor were labeled by CD34, then the microvessel density (MVD) was calculated. Results The expression of VEGF and bFGF in hemangioblastoma was significantly higher than those in normal brain tissue(P 〈 O. 01 ). MVD was significantly higher in hemangioblastoma than that in normal brain tissue (P 〈 0.01 ). There was positive correlation between the expression of VEGF and bFGF ( r = 0. 541 ,P 〈 0.01 ). But there was no significant difference between the cystic and solid tumors on the expression level of VEGF, bFGF and MVD (P 〉 0.05 ). Conclusion here may be a causal relationship between VEGF and bFGF in mechanism. Examination of bFGF and VEGF may provide a new thought on the pathogenesis and treatment of hemangioblastoma. And anti-VEGF and anti-hFGF therapy would be a new treatment tbr hemangioblastoma.
