摘要 目的探讨人参皂苷Rg3对人宫颈癌Hela细胞的诱导凋亡作用。方法用浓度梯度Rg3处理Hela细胞,用流式细胞仪方法检测细胞凋亡及细胞周期的变化,用RT-PCR法检测凋亡相关基因Bcl-2、bax表达。结果 10、20、40 mg/L Rg3作用Hela细胞24 h后的凋亡率分别为(5.6±1.4)%、(10.3±2.7)%、(21.2±4.9)%,均高于对照组(P<0.01);同时随着Rg3浓度的增加,G2/M期细胞比例升高,Bcl-2表达明显降低,bax基因表达明显升高(P<0.01)。结论 Rg3能将Hela细胞阻滞在G2/M期,通过上调bax和下调Bcl-2表达诱导肿瘤细胞凋亡。其作用呈浓度依赖性。 Objective To investigate the apoptotic effect of ginsenoside Rg3 on Hela cells.Methods After treated with ginsenoside Rg3,Hela cell apoptosis and cell cycle were detected by cell flow cytometry(FCM).The expression of Bcl-2/bax mRNA was detected by RT-PCR.Results At dosage of 10,20 and 40 mg/L for 24 h,Hela cell apoptosis raised to(5.6±1.4)%,(10.3±2.7)% and(21.2±4.9)%,respectively(P0.01).After treated with Rg3,the cell cycle of treated Hela cells showed increased percentage of G2/M stage maintenance.At the same time,RT-PCR showed that Rg3 down-regulated the expression of Bcl-2 mRNA and up-regulated that of bax mRNA(P0.01).Conclusion Rg3 can inhibit the cell cycle at G2/M phase and induce apoptosis in the Hela cells by down-regulating Bcl-2 mRNA and up-regulating bax mRNA in a dose-dependent manner.
