摘要
目的:探讨黄芪多糖(APS)对60Coγ射线损伤大鼠睾丸间质细胞的防治作用。方法:雄性SD大鼠30只,随机分为空白对照组、60Coγ照射组、APS+60Coγ照射组。观察各组大鼠睾丸间质细胞的形态学改变;ELISA法检测血清睾酮(T)、黄体生成素(LH)的含量;RT-PCR法检测睾丸间质细胞黄体生成素受体(LHR)mRNA水平。结果:与空白对照组相比,60Coγ照射组大鼠睾丸间质细胞形态发生改变,细胞体积缩小,胞质减少,细胞核皱缩,数目减少;APS+60Coγ照射组睾丸组织病理改变较60Coγ照射组减轻。3组大鼠血清T及LH水平差异无统计学意义(F=2.446和1.142,P>0.05),睾丸组织中LHR的mRNA的表达差异有统计学意义(F=24.032,P<0.001)。结论:APS可防治60Coγ射线对睾丸间质细胞损伤,其机制可能与促进睾丸间质细胞LHRmRNA的高表达有关。
Aim:To investigate the protection of astragalus polysacharin(APS) on the impairment of the leydig cells caused by higher doses of 60Co γ in rats.Methods:Thirty male SD rats were randomized into three groups(n=10):the normal control group,the radiation control group,the APS plus radiation group.The structural changes of the leydig cells were observed with light microscope.The serum luteinizing hormone(LH) and testosterone(T) levels were measured by ELISA;and the mRNA expression level of luteinizing hormone receptor(LHR) from the leydig cells surface was measured by RT-PCR.Results:Compared with the normal control group,the leydig cells exhibited decreases in size,cytoplasm,and quantity as well as the nuclear shrinkage after radiation.The mRNA expression levels of LHR were lower(P0.01);in the APS irrigation group,the pathological changes of leydig cells were lightly.The mRNA expression levels were close to the normal;the serum LH and T levels were no significant change.Conclusion:APS can reduce the impairment of the leydig cells caused by the higher doses 60Co γ ray in rats,which may be involved in its induce the high expression of LHR mRNA.
