大孔树脂与氧化铝联用纯化南五味子总木脂素的工艺优选 被引量:6

Optimization of Purification Technology of Total Lignans from Schisandra sphenanthera in Conjunction with Macroporous Resin and Aluminum Oxide Column

摘要 目的:优选大孔树脂与氧化铝联用对南五味子总木脂素提取液的纯化工艺。方法:以除杂率和五味子酯甲转移率的综合评分为指标,采用正交试验考察树脂用量、乙醇体积分数和乙醇用量对南五味子总木脂素大孔树脂纯化工艺的影响;通过单因素试验考察南五味子总木脂素的氧化铝吸附除杂工艺。采用紫外分光光度法测定总木脂素含量,HPLC测定五味子酯甲、五味子甲素的含量。结果:最佳纯化工艺为取2/3~4/3倍量LSA-7型大孔树脂,加85%乙醇6 BV洗脱,洗脱液过1倍药材量氧化铝柱吸附除杂。结论:该优选工艺合理、稳定可行,适用于工业生产。 Objective: To optimize purification technology of total lagnans from Schisandra sphenanthera extract by macroporous resin combined with aluminum oxide. Method: With composite score of impurity removal rate and transfer rate of schisantherin A as index, effect of the amount of resin, ethanol concentration and amount on macroporous resin purification technology of total lignans was investigated by L9 (34) orthogonal test; alumina adsorption purifying technology of total lignans from S. sphenanthera was investigated by single factor test. Thecontent of total lignans was determined UV spectrophotometry, HPLC was used to determine the contents of schisantherin A and deoxyschizandrin. Result: Optimum purification conditions of total ligans in S. sphenanthera were as follows: took 2/5-4/3 times the amount of LSA-7 macroporous resin, eluted with 6 BV 85% ethanol, ratio of aluminum oxide and crude drug 1: 1. Conclusion: This optimized purification process was feasible, stable, reasonable and suitable for industrial production.
