反相高效液相色谱法测定丁香挥发油中丁香酚含量

摘要 目的建立反相高效液相色谱法,测定丁香挥发油中丁香酚的含量。方法采用Lichrospher C18色谱柱(150mm×4.6mm,5μm),流动相为甲醇-水(64:36),柱温25℃,流速为1.0ml/min,检测波长280nm,以外标法峰面积定量。结果丁香酚与挥发油中其他成分的分离效果良好,在0.11~110.00mg/L浓度范围内呈良好的线性关系,回归方程为:Y=17.74C+1.518(r=0.9999,n=5);平均回收率为100.06%,相对标准偏差为0.16%。结论本方法简便、准确,可快速测定丁香挥发油中丁香酚的含量,可为质量控制提供依据。 Objective To establish a reversed-phase high performance liquid chromatography (RP-HPLC) for simultaneous determination of eugenol in the essential oil from Clove. Method The mobile phase was carbinolwater(64: 36) and the flow rate was about 1.0 ml/min. Lichrospher C18 (150 mm ×4.6 mm, 5 μm) column was used and the detective wavelength was 280 nm. Results The calibration curves were linear in the concentration range of 0.11 -110.00 mg/L for eugenol. The regression equation was Y = 17.74 C + 1. 518 ( r = 0. 9999, n = 5 ) , the average recovery was 100.06% and the relative standard deviations was 0.16%. Conclusion Our established method is simple, quick and accurate for the simultaneous separation and determination of eugenol in the essential oil from Clove.
