摘要
目的:研究左归丸对谷氨酸(Glutamic acid)诱导的小鼠神经毒性的可能保护作用。方法:除对照组外,其他组小鼠接受谷氨酸单钠(monosodium glutamate,MSG,4.0g/kg)灌胃,每日一次,连续给药10日。第11天起,治疗组分别接受左归丸(1.0,2.0,4.0 g/kg,ig),之后通过小鼠的行为学实验、脑海马组织病理学和脑组织抗氧化指标检测,分析MSG所引起的功能和形态学上的变化以及左归丸的可能的保护作用。结果:左归丸显著修复MSG对小鼠行为、脑海马组织病理的不良影响,明显升高小鼠脑组织中超氧化物歧化酶(SOD)水平,而显著降低丙二醛(MDA)。结论:左归丸对MSG诱导的小鼠神经毒性有一定的保护作用,其保护作用机制可能与其改善小鼠抗氧化指标水平,增强机体的抗氧化能力有关。
To investigate a possible protective effect of Zuogni Pill(ZGW) on glutamate-induced neurotoxicity in adult mice. Methods: Mice, except controls, received intragastric (ig) adminstration of monosodium glutamate (MSG , 4.0g/kg/d) for 10 days, at the l lth day, each mouse received ig adminstration of ZGW (1.0, 2.0, 4.0 g/kg/d, ig) or physiological saline for 30 days. The mice in controls received ig administraion of physiological saline instead of ZGW. and then behavioral tests, histopathological examinations of hippocampi of brains were used, the activity of SOD and the concentration of MDA in cerebral tissue in mice were measured by the method of biochemistry and mo- lecularbiology to analyze glutamate (Glu)-induced functional, morphological, and molecular cell biological changes of the brains and a possi- ble protection of ZGW. Results: The results showed that administration of ZGW obviously facilitated brain repair of glutamate-induced behav- ioral disturbances and histopathulogieal impairs in mice; and the activity of SOD of ZGW groups increased significantly ( P 〈 0.05 or P 〈 0. 01 ) in cerebral tissue, while the concentration of MDA decreased significantly (P 〈0.05) compared with Model group. Conclusion: ZGW has neuroprotection against glutamate-induced neurotoxieity in mice, and its effectivem echanism may be associated with antioxidant ability enhanced by ZGW in mice.
