左归丸对痴呆鼠抗氧化作用及尼氏体的影响 被引量:8

摘要 目的探讨左归丸对痴呆大鼠抗氧化作用及尼氏体的影响。方法将84只Wistar雄性大鼠随机分成6组:A组腹腔注射生理盐水再脑内海马注射生理盐水,B、C、D、E、F组以D-半乳糖腹腔注射合并β淀粉样蛋白1-40注射海马制备老年性痴呆(AD)动物模型,C、D、E、F组同时分别灌胃抗脑衰胶囊、哈伯因和左归丸[5.4和2.7g/(kg.d)]。采用生化法检测各组大鼠脑匀浆过氧化氢酶(CAT)、单胺氧化酶(MAO),尼氏染色观察各组大鼠海马神经元尼氏体及电镜观察各组超微结构变化。结果灌服后左归丸组均能明显提高AD大鼠脑组织CAT活性、抑制MAO活性(P<0.05);左归丸治疗组尼氏体溶解明显减少;F组神经纤维结构完好,尼氏体丰富,突触结构尚可。结论左归丸通过提高抗氧化能力和改善神经元细胞损害对AD大鼠有一定的防治作用。 Objective To investigate the effects of Zuoguiwan on antioxidation and Nissl' s body of Alzheimer' s disease (AD) model rats. Methods Totally 84 Wistar rats 'were randomly divided into 6 groups. Sodium chloride was injected into hippocampus after injected into abdominal cavity of the rats of group A. The abdominal cavity were injected by Dgalactose in groups B, C, D, E and F and β amyloid 1-40 was injected into hippocampus to make Alzheimer' s disease (AD) model rats. Groups C and D were treated by Kang Nao Shuai Capsule and Huperzine A, respectively. Groups E and F were fed of Zuoguiwan with 5.4 and 2.7 g/( kg . d) , respectively. The contents of CAT and MAO in the brain were measured by biochemistry. Uhrastructure was observed by electron microscope and Nissl' s body were observed by Nissl' s staining. Results Zuoguiwan could reduce the activity of MAO and improve the activity of CAT in the brain ( P 〈 0.05 ). Zuoguiwan could reduce the solution of Nissl' s body in the hippoeampus. The structure of nerve fibre in the Zuoguiwan 5.4 g/kg per day group was intact with the plenty Nissl' s bodys and the well structure of synapse. Conclusion Zuoguiwan may have preventive and therapeutic effects through improving antioxidation activity and cell' s injury on AD model rats.
