姜黄素对骨关节炎软骨细胞增殖及分泌MMP-13,IL-6的影响 被引量:32

摘要 目的研究姜黄素对骨关节炎(OA)患者软骨细胞增殖、细胞因子MMP-13和IL-6分泌的影响。方法收集4例OA患者关节软骨作为实验组,3例股骨头骨折的关节软骨为对照组,分离软骨细胞进行体外培养。使用浓度为0,10,40,80,120μmol/L的姜黄素溶液共培养软骨细胞48 h,MTT检测姜黄素对软骨细胞增殖的作用,ELISA检测共培养细胞上清液中MMP-13和IL-6含量。结果姜黄素在40μmol/L浓度以上对OA软骨细胞增殖具有明显抑制作用(P<0.05),120μmol/L浓度对正常软骨细胞有明显抑制作用(P<0.05);80μmol/L以上浓度对OA软骨细胞和正常软骨细胞分泌MMP-13具有抑制作用(P<0.05);80μmol/L以上浓度对OA软骨细胞分泌IL-6具有抑制作用(P<0.05),120μmol/L浓度时对正常软骨细胞分泌IL-6具有抑制作用(P<0.05)。结论姜黄素能够抑制软骨细胞的细胞增殖,抑制软骨细胞释放MMP-13和IL-6,减轻炎症反应,保护软骨细胞,延缓软骨退变。 Objective It is to study the effect of curcumin on the cells proliferation and the secretion of MMP- 13 ,IL- 6 of chondrocyte in osteoarthritis (OA). Methods Articular cartilage of 4 cases of patients with OA was collected as experimental group, that of 3 cases with femoral head fracture was collected as control group. The chondrocyte were cultured in vitro with curcumin ( 0,10,40,80 and 120μmol/L) for 48 h. The proliferation of chondrocytes were analyzed by MTT. The concentra- tion of MMP - 13 and IL - 6 was measured by ELISA. Results The curcumin could inhibit obviously the proliferation of OA chondroeyte when its concentration was more than 40 μmol/L( P 〈 0.05 ) , and the normal chondrocyte could be inhibited when the concentration was 120 μmol/L(P 〈0.05). When its concentration was more than 80 p.mol/L it could inhibit the section of MMP - 13 in OA and normal chondrocyte( P 〈0. 05 ). The IL -6 in OA ehondrocyte was inhibited when the concentration was more than 80 p.mol/L(P 〈 0.05 ), and the normal chondrocyte was inhibited when the concentration wasl20 p^mol/L( P 〈 0.05). Conclusion Curcumin can inhibit the secretion of MMP -13 and IL -6, and inhibit the ehondrocyte proliferation, thus to relieve inflammation, protect chondrocyte and delay cartilagedegeneration.
