摘要
目的了解广西柳江县农村大肠杆菌的耐药性及耐药性大肠杆菌gyrA基因突变特点。方法收集广西柳江县农村腹痛、腹泻患者的粪便,分离培养大肠杆菌,采用K-B法测菌株的耐药性;PCR扩增对氧氟沙星、环氧沙星耐药菌株的gyrA基因,检测gyrA基因序列,分析突变位点。结果经鉴定获得的312株大肠杆菌中,对氧氟沙星、环丙沙星耐药的分别有206株和179株,耐药率分别为66.0%和57.4%,所有耐药株均出现gyrA基因突变,主要是第83和87位点分别出现C-T和G-A的突变。结论柳江县农村患者分离的大肠杆菌对喹诺酮类药物耐药性明显,耐药与gyrA基因突变有密切的关系,基因突变促使丝氨酸和天冬氨酸分别变为亮氨酸和天冬酞胺。
Objective To explore the characteristics of drug resistance and gyrA gene mutation of Escherichia coli in the rural area,Liujiang County of Guangxi. Methods The stool from the patients with abdominal pain and diarrhea in the rural area,Liujiang County of Guangxi ,was collected. Escherichia coli was isolated and cultured,and strain resistance was measured by K-B method. The strains of gyrA gene resistant to ofloxacin and cipmfloxacin were amplified by PCR. The sequence of gyrA gene was detected and its mutation was analyzed. Results In 312 strains of Escherichia coli which had been identified ,206 strains were resistant to ofloxacin and 179 stains resistant to ciprofloxacin ,the resistance rates were 66.0% and 57.4% ,respectively. AU strains showed gyrA gene mutation,mainly C-T mutation in the 83rd locus and G-A mutation in the 87th locus. Conclusion Escherichia coli isolated from the patients in the rural area, Liujiang County of Guangxi,is resistant to quinolones significantly, and its resistance is closely correlated with gyrA gene mutation. Serine becomes leucine and aspartic acid becomes Asx induced by the gene mutation.
