摘要
目的研究Toll样受体3(TLR3)在糖尿病模型大鼠脊髓中的表达,探讨其与糖尿病神经病理性疼痛的关系。方法 50只SD大鼠,其中40只大鼠一次性腹腔注射链脲佐菌素(STZ)65mg/kg建立糖尿病大鼠模型,C组腹腔注射相同剂量的柠檬酸/柠檬酸钠缓冲液。1周后断尾取血测血糖,空腹血糖>16.7mmol/L为糖尿病模型制备成功。分别测定大鼠制模后2周(D2组)、4周(D4组)、6周(D6组)、8周(D8组)机械痛阈(MWT)和热痛阈(TWL)。采用RT-PCR法测定大鼠L4~6脊髓TLR3表达和TNF-α水平。结果与C组比较,D2、D4、D6、D8组大鼠MWT明显降低(P<0.05),脊髓TLR3表达、TNF-α水平明显升高(P<0.05);与D2组比较,D4、D6、D8组大鼠MWT明显降低(P<0.05),TLR3表达、TNF-α水平明显升高(P<0.05)。与C组和D2组比较,D4、D6、D8组大鼠TWL明显降低(P<0.05)。结论糖尿病大鼠脊髓TLR3表达出现上调,并可能通过促使TNF-α释放诱发神经病理性疼痛。
Objective To observe the expression of toll-like receptor 3 (TLR3) in the spinal cord of diabetic rats and its correlation with diabetic neuropathic pain. Methods Fifty male SD rats were divided into diabetic model group (group D, n=40) and control group (group C, n=10). Rats in group D were intraperitoneally injected with streptozotocin 65 mg/kg to establish the diabetic model, while rats in group C were injected with citric acid/sodium citrate. Mechanical withdrawl threshold (MWT) and thermal withdrawl latency (TWL) were measured in C group and D group at two weeks (group I)2), four weeks (group D4), six weeks (group DG ), and eight weeks (group Ds) after the model established, TLR3 expression and TNF-a level of the L46 spinal cord of rats were detected. Results MWT was lower and TLR3 expression and TNF-a level were higher in group D than in group C (P〈0. 05). Compared with group 132, MWT in group D4, D6, and D8 was lower and TLR3 expression and TNF-a level were higher (P〈0. 05). Compared with groups D2 and C, TWL decreased in groups D4, D6, and D8 (P〈0.05). Conclusion TLR3 expression was higher in diabetic rat spinal cord, which might induce neuropathic pain via the increased TNF-a release.
