摘要
目的:筛选参芎化瘀胶囊对治疗全脑缺血再灌注损伤大鼠的推荐使用剂量,为提高临床疗效奠定基础。方法:成年健康雄性SD大鼠110只随机分为假手术组、模型组及参芎化瘀胶囊低、中、高剂量组(240,360,480 mg.kg-1),改良的Pulsineli 4血管阻断(4-VO)法制作全脑缺血模型,术后24,48,72 h苏木精-伊红(HE)染色观察各组脑组织病理学形态的变化;72 h后应用横木行走实验(BWT)和抓握力量测试评估并记录感觉运动整合功能水平;选择最佳剂量组应用免疫组化法检测血管内皮生长因子(VEGF)的表达。结果:与假手术组比较,光镜下观察脑缺血后神经元内细胞形态明显受损伤,神经元密度减少,感觉运动整合功能水平明显降低(P<0.01);与模型组比较,参芎化瘀胶囊组神经元密度以及感觉运动检测评分均高于模型组(P<0.05),上述变化在高剂量组最为明显。选择高剂量组进行免疫组化检测VEGF,假手术组呈较低水平表达;与假手术组比较,模型组表达明显增加(P<0.05),参芎化瘀胶囊用药干预组VEGF表达进一步增加(P<0.01)。结论:参芎化瘀胶囊对大鼠脑缺血再灌注损伤有治疗作用,推荐剂量是480 mg.kg-1,其疗效机制可能与促进VEGF表达有关。
Objective： To select the recommended dose of the Shenxiong Huayu capsule in the treatment of global cerebral ischemia/reperfusion injury in rats. Method： Adult male SD rats were divided into five groups randomly： sham group, model group, low, medium and high dose group of Shenxiong Huayu capsule group （240, 360, 480 mg .kg-1）. Global cerebral ischemia model was formed by improved four-vessel occlusion as described according to the description of Pulsinelli＇s method. Morphological changes were observed by HE staining in 12, 24, 48 h after injury. Beam walking test and bilateral forepaws grasp was used to detecte sensorimotor integration after 72 h. The best dose group was selected by using immunohistochemlstry method to detect the expression of vascular endothelial growth factor （VEGF）. Result： Compared with sham group, neuronal morphous was damaged, the density of neurons was reduced, the level of sensorimotor integration was decreased obviously （P 〈 0. 01 ） in model group Compared with model group, the number of neurons and the level of sensorimotor integration were increased obviously in Shenxiong Huayu capsule group （P 〈 0.05） ; the above mentioned changes were more significant in high dose of Shenxiong Huayu capsule group. The expression of VEGF in high dose group was increased significantly, compared with model group （P 〈 0. 01 ）. Conclusion： Shenxiong Huayu capsule have good therapeutic effect on ischemia-reperfusion injury and the recommended dose is 480 mg ·kg-1, which is related to regulating VEGF expression.
