摘要
【目的】建立人参皂苷Rg1、三七皂苷R1经皮给药微透析采样的体外回收率研究方法。【方法】运用高效液相色谱法测定微透析样品中人参皂苷Rg1、三七皂苷R1的浓度,利用增量法、减量法计算体外回收率,并考察流速、浓度、温度对回收率的影响。【结果】在同一浓度下,体外回收率随着流速的增加而减少,采用1μL/min流速灌流,30 min取样时间间隔的参数适于人参皂苷Rg1、三七皂苷R1的微透析研究;在同一流速下,浓度的变化对体外回收率的影响不大;体外回收率随温度的上升明显增加。【结论】确定了影响微透析体外回收率的因素,建立了人参皂苷Rg1、三七皂苷R1经皮给药微透析体外回收率的研究方法。
Objective To establish a method for detecting the in-vitro recovery rate of ginsenoside Rg 1 and notoginsenoside R 1 in microdialysis probe through transdermal administration.Methods The concentration of ginsenoside Rg 1 and notoginsenoside R 1 in microdialysate samples was detected by high performance liquid chromatography(HPLC).The in-vitro recoveries were obtained through recovery estimated by increment method and decrement method.The effect of flow rates,sample concentrations and temperature on the recovery was also studied.Results At the same concentration,in-vitro recovery rate decreased with the increase of flow rate,and the optimal conditions for ginsenoside Rg 1 and notoginsenoside R 1 microdialysis were perfusion at flow rate of 1μL / min and sampling every 30 min.At the same flow rate,the concentration had no obvious effect on in-vitro recovery.Invitro recovery rate increased significantly with the increase of temperature.Conclusion The influencing factors of in-vitro recovery rate in microdialysis probe have been worked out,and a retrodialysis method has been established for the detection of in-vitro recovery rate of ginsenoside Rg 1 and notoginsenoside R 1.
