摘要
文章合成了奥美拉唑,并用光学拆分剂拆分得到S-奥美拉唑即埃索美拉唑,拆分条件为S-(-)-1,1'-联萘-2,2'-二酚、外消旋奥美拉唑和碱的物质的量比为1∶1.6∶1.8,乙醇和水的体积比为3.7∶1,室温反应12 h,埃索美拉唑的镁盐为89%的产率和97.95%的ee值。
Omeprazole was synthesized and resolved by chiral resolution agent to obtain the S-isomer (Esomeprazole). The resolving conditions were as follows: S -(-)-1 1'-Binaphthyl-2,2'-Diol, racemic omeprazole and alkali (molar ratio 1 : 1.6: 1.8) reacted in ethanol and water (3.7 : 1 in v : v ) at room temperature for 12 h. The magnesium salt of Esomeprazole was obtained in a high yield (89 %) and excellent enantioselectivity (97.95 % ee).
