我国百日咳疫苗生产用菌株主要抗原片段基因序列分析 被引量:4

摘要 目的对我国百日咳疫苗生产用菌株主要抗原片段基因进行序列分析,为我国百日咳疫苗的研制和质量控制提供依据。方法分别从我国百日咳杆菌疫苗生产用菌株CS株基因组DNA中扩增百日咳毒素(Pertussis toxin,PT)、外膜蛋白(Pertactin,PRN)、菌毛蛋白2(Fimbriae 2,FIM2)、菌毛蛋白3(Fimbriae 3,FIM3)基因,克隆至pMD18-T载体,测序,并与GenBank中登录的百日咳杆菌其他菌株的PT、PRN、FIM2、FIM3基因序列进行比较,构建系统进化树。结果百日咳杆菌CS株PT、PRN、FIM2和FIM3基因的扩增产物大小均与预期一致。CS株PT基因与国内的另一疫苗生产株18323亲缘关系最近,在同一分支上,且与国际标准株Tohama在同一亚分支内;PRN、FIM2和FIM3基因与国际标准株Tohama均在同一进化分支上。结论百日咳杆菌CS株与国际标准菌株Tohama亲缘关系较近,与临床分离的其他流行株亲缘关系较远。 Objective To analysis the gene sequence of major antigen of pertussis vaccine strain in China.Methods The pertussis toxin(PT),pertactin(PRN),fimbriae 2(FIM2)and FIM3 genes of CS strain for production of pertussis vaccine were amplified by PCR and cloned into pMD-18T vector for sequencing,and the results were compared with those of other strains reported in GenBank,based on which a phylogenetic tree was plotted.Results The lengths of amplified PT,PRN,FIM2 and FIM3 genes were consistent with those expected.The PT gene of CS strain belonged to the same branch of another vaccine strain 18323 in China,and the same sub-branch of international standard Tohama strain.However,the PRN,FIM2 and FIM3 genes belonged to the same branch of Tohama strain.Conclusion CS strain showed close relationship to international standard Tohama strain,while showed far relationship to other clinical isolates,on phylogenetic tree.
