瞬时受体电位通道蛋白A1和V1在正常小鼠耳蜗中的定位表达 被引量:1

摘要 目的研究瞬时受体电位通道蛋白A1(TRPA1)和V1(TRPV1)在正常小鼠耳蜗中的定位与表达。方法选择12只健康成年BALB/c小鼠,应用免疫荧光染色方法和蛋白质印迹技术检测TRPA1和TRPV1在耳蜗中的定位与表达,同时结合电生理指标听脑干反应(auditory brainstem response,ABR)测试小鼠的听力。结果 TRPA1和TRPV1在小鼠耳蜗螺旋器的内、外毛细胞、螺旋神经节细胞和血管纹边缘细胞等都有表达,且主要定位于细胞膜;蛋白质印迹检测结果亦显示小鼠耳蜗中有TRPA1和TRPV1的表达。结论 TRPA1和TRPV1在正常BALB/c小鼠耳蜗的多种细胞中均有表达,这为深入研究二者在内耳听觉生理和病理生理中的作用奠定基础。 【Objective】 To investigate the localization and expression of transient receptor potential A1(TRPA1) and transient receptor potential vanilloid 1(TRPV1) in normal mouse cochlea.【Methods】 12 healthy adult BALB/c mice were used for the experiment,TRPA1 and TRPV1's localization and expression in the normal mouse cochlea were detected by the immunofluorescence staining and western blot.【Results】 TRPA1 and TRPV1 were expressed in inner and outer hair cells of the organ of corti,and spiral ganglion cells and marginal cells of stria vascularis.Moreover,TRPA1 and TRPV1 were located in the cytoplasm and plasma membrane.The result of western blot also revealed the expression of TRPA1 and TRPV1 in normal mouse cochlea.【Conclusions】 TRPA1 and TRPV1 were expressed in many cell types of normal mouse cochlea,which settles firm base for intensive studying the role of TRPA1 and TRPV1 in auditory physiology and pathophysiology of inner ear.
