摘要 研究鹿角盘中蛋白质酶法水解条件。用胰酶、胰蛋白酶、木瓜蛋白酶在相同条件下水解鹿角盘蛋白质,比较氨基氮生成率,选择适宜酶类。用所选定的酶在不同温度(T)、不同底物浓度比(E/S)和不同水解时间(t)水解鹿角盘蛋白质,以氨基氮生成率为指标确定最佳水解条件。胰蛋白酶为适宜酶,最佳水解条件为:温度45℃,pH 8.0,底物浓度7.5%,酶与底物浓度比6 000μ/g,时间6h。胰蛋白酶是水解鹿角盘蛋白质较适宜的酶,在上述最佳条件下,水解鹿角盘蛋白质,氨基氮生成率达42%,氮溶解指数达93%,水解液中蛋白质绝大多数为低肽分子,更有利于机体消化吸收。 To study protein enzymatic hydrolysis conditions from antler plate. Antler plate protein was hydrolyzed with trypsin, pancreatin and pa- pain under the same condition, amino nitrogen production rate was eompared, suitable enzyme was choosed. The protein enzymatic hydrolysis in antler plate was researched with different temperature, enzyme/substrate ratio, and hydrolysis time. The optimum parameters of reaction were de- termined with amino nitrogen as the index. Trypsin is suitable for protein hydrolysis in antler plate. The results showed that the optimum condi- tions of hydrolysis are the temperature at 45℃, the original pH 8.0, substrate concentration 7.5 %, enzyme/substrate ratio( E/S)6 000μ/g, and hydrolysis time 6h. Trypsin is suitable enzyme for protein hydrolysis in antler plate. Under optimum conditions, amino nitrogen production rate is up to 42% ,the index of soluble nitrogen is up to 93% in hydrolysate. The most of protein in hydrolysate is low molecular weight peptide, which is good for digestion and absorption of human body.
