摘要
[目的]利用醋酸纤维素薄膜电泳分析金针菇酯酶同工酶。[方法]以6种金针菇为材料,采用醋酸纤维素薄膜电泳对其酯酶同工酶进行分析,并与利用PAGE法得到的针菇酯酶同工酶图谱进行对比。[结果]6种金针菇共电泳得到20条酯酶同工酶带,其中相对迁移率不同的共有8条,大小分别为0.580、0.613、0.645、0.709、0.806、0.839、0.871、0.934;6种金针菇中,①号和②号有3条相同的带,④号和⑤号条带完全相同,③号与④号、⑤号有2条相同的带,⑥号与其他5种菌株间相同的条带较少。[结论]该试验结果与PAGE法显示的金针菇酯酶同工酶图谱基本一致,为实现利用醋酸纤维素薄膜电泳技术快速检测各类食用菌酯酶同工酶提供了可能。
[Objective] To analyze the esterase isozymes of Flammulina velutipes by cellulose acetate membrance electrophoresis.[Method] The esterase isozymes of six kinds of Flammulina velutipes were analyzed by cellulose acetate membrance electrophoresis and the result was compared with esterase isoenzyme PAGE patterns.[Result] The electrophoretic six kinds of Flammulina velutipes were obtained 20 esterase isozyme bands,in which 8 bands had different relative mobility,the size was 0.580,0.613,0.645,0.709,0.806,0.839,0.871,0.934 respectively;in the six kinds of Flammulina velutipes,No.1 and No.2 had 3 same bands,No.4 and No.5 had the same bands,No.3 had 2 bands the same with No.4 and No.5,No.6 had little same bands with the other 5 strains.[Conclusion] The test results and the PAGE patterns of Flammulina velutipes esterase isozymes were basically the same,and it provides the possible for the realization of using cellulose acetate membrane electrophoresis technique for esterase isozymes rapid detection of various types of domestic fungus.
