摘要
目的观察甘肃产藏药五脉绿绒蒿有效部位对肝纤维化大鼠炎症相关因子的影响。方法在给药的基础上,采用皮下注射CCl4、喂饲高脂低蛋白复合饲料并饮用20%乙醇来复制肝纤维化动物模型,6周后对大鼠肝组织HE染色,做病理组织学观察,检测血清酶与相关细胞因子水平,对甘肃产藏药五脉绿绒蒿各提取部位防治肝纤维化疗效进行评价。结果复合因素肝纤维化造模结束后,模型对照组大鼠肝组织病理结果显示其肝纤维化处于Ⅱ-Ⅲ期;与模型对照组比较,甘肃产藏药五脉绿绒蒿醇提物、黄碱组以及总黄酮能明显降低血清ALT、AST与TNF-α、IL-6,显著升高IFN-γ,改善纤维化大鼠细胞因子水平失衡状态,同时发现总黄酮的作用有一定的量效关系。结论甘肃产藏药五脉绿绒蒿醇提物及其中的总黄酮对CCl4复合因素诱导的肝纤维化大鼠血清细胞因子水平失衡有一定的改善作用。
To investigate the influence of active fractions of Meconopsis quintuplinervia Regel (MQR) growing in Gansu province on related cytokines of experimental liver fibrosis in rat. By subcutaneous injection of CCl4, feeding high-fat low-protein diet and drinking 20% alcohol for 6 weeks, the model of hepatic fibrosis rats was established to evaluate the effects of the extractive parts from MQR through detecting serum cytokines and observing hepatic pathology. We found that the ethanol extractive, total flavones, and the mixture of total flavones and total alkaloids from MQR growing in Gansu province could significantly decrease ALT, AST, TNF-α, and IL-6 in serum, but increase IFN-γ and improve disequilibrium of cytokines in experimental liver fibrosis rat. Furthermore, the effects of total flavones exhibited dose-effect relationship. All the result suggested that the ethanol extractive and total flavones of MQR growing in Gansu province have better balancing effects on disequilibrium of cytokines of liver fibrosis in rats induced by CCl4 composited factor.
