“标本配穴”针刺对糖尿病大鼠下丘脑瘦素受体及其mRNA表达的影响 被引量:12

Effect of Acupuncture Therapy on the Expressions of LeptinR and Its mRNA in the Hypothalamus of STZ-induced Diabetic Rats

摘要 目的:观测针刺对糖尿病大鼠下丘脑Leptin受体及其mRNA表达的影响。方法:180~220gWistar大鼠43只,13只为正常对照,另30只行链脲左菌素腹腔注射造模,造模成功的大鼠随机分为模型组和针刺治疗组(针刺胰俞、足三里、关元),针刺治疗2个疗程后取脑,运用免疫组化法和原位杂交法分别检测下丘脑LeptinR蛋白及其mRNA的表达。结果:STZ糖尿病大鼠下丘脑室旁核、弓状核LeptinR免疫阳性物较正常组大鼠明显减少,下丘脑外侧区LeptinRmRNA表达较正常明显减少(P<0.05),针刺治疗后糖尿病大鼠下丘脑室旁核、弓状核LeptinR及下丘脑外侧区LeptinRmRNA表达明显增多(P<0.05)。结论:"标本配穴"针刺能显著升高糖尿病大鼠下丘脑低水平的Leptin受体及其基因的表达,有效发挥其调节体重、改善胰岛素敏感性和葡萄糖代谢的作用。这可能是针刺调节糖尿病大鼠内分泌的重要中枢机制之一。 Objective : To observe the effect of acupuncture therapy on the expressions of LeptinR and its mRNA in the hypothalamus of STZ - induced diabetic rats. Methods : Among forty - three 180 ~220g Wistar rats, thirteen were assigned as normal control group and thirty were induced diabetes by injecting STZ into celiac cavity. The diabetic rats were ran domly divided into model group and treatment group( needling Yishu, RN4 and ST36). After two courses of treatment,the rats were sacrificed to test the expression of LeptinR and its mRNA in the hypothalamus with immunohistochemical and in situ hybridization methods respectively. Results :Compared with the normal group, the concentration of LeptinR in hypo- thalamic paraventricular nucleus (PVN) and arcuate nucleus (ARC) and expression of its mRNA in lateral hypothalamus (LH) were decreased significantly in STZ - induced diabetic rats. After acupuncture, the concentration of LeptinR and ex pression of its mRNA in hypothalamns were increased obviously. Conclusion :Acupuncture can increase the low concen tration of LeptinR and its synthesis in hypothalamus of STZ - induced diabetic rats ,which may be one of the central mech anisms of acupuncture in regulating energy metabolism of diabetes.
