摘要
目的观察miR-181a对A549/DDP细胞顺铂敏感性的影响,并探讨其机制。方法采用miR-181a mimic转染A549/DDP细胞,MTT法检测细胞增长率;荧光定量PCR检测A549/DDPmiR-181a表达;免疫印迹法检测Bcl-2蛋白表达;AnnexinV/PI双标法检测细胞凋亡。结果 miR-181a提高A549/DDP细胞的顺铂敏感性,促进顺铂诱导的细胞凋亡,并且降低Bcl-2蛋白的表达。结论 miR-181a通过降低Bcl-2蛋白表达,促进顺铂诱导的细胞凋亡,从而提高A549/DDP细胞顺铂敏感性。
Objective To investigate the effect of miR-181a on the sensitivity of A549/DDP cells to cisplatin and its mechanism.Methods A549/DDP cells were transfected with miR-181a mimic.The cell growth was detected by MTT method.The expression of microRNA-181a was measured by fluorescence quantitative polymerase chain reaction.Bcl-2 protein level was determined by Western blotting.The apoptosis of cells was measured by Annexin V/PI methods.Results miR-181a significantly enhanced the sensitivity of A549/DDP cells to cisplatin and the apoptosis induced by cisplatin.After transfected with microRNA-181a mimic,Bcl-2 protein level decreased in A549/DDP cells.Conclusions MicroRNA-181a can enhance the sensitivity of A549/DDP cells to cisplatin,which may be associated with down-regulation of Bcl-2 expression.
