摘要
目的探讨抗凋亡基因Bag-1和Caspase-3在正常大肠黏膜和大肠癌中的表达变化及与大肠癌发生、发展和转移的关系。方法采用免疫组化SP法,分别检测15例正常大肠黏膜和66例大肠癌中Bag-1和Caspase-3蛋白表达情况。结果 Bag-1蛋白在正常肠黏膜、大肠癌中的阳性表达率分别为13.3%、74.2%。Caspase-3蛋白在正常大肠黏膜和大肠癌中的阳性表达率分别为46.7%、68.2%。Bag-1蛋白表达与大肠癌分化程度、Duke's分期及淋巴结转移有关(P<0.05),而Caspase-3蛋白表达与分化程度、Duke's分期及淋巴结转移无关(P>0.05)。大肠癌中Bag-1的表达与Caspase-3明显相关(P<0.05)。结论 Bag-1和Caspase-3参与了大肠癌的发生发展过程,Bag-1可作为判断大肠癌生物学行为的新的生物学指标。
Objective To investigate the relationship and clinical significance of Bag-1 and Caspase-3 expression in colorectal carcinoma tissues and normal colorectal tissues.Methods SP immunohistochemical staining was used to observe the expression of Bag-1 and Caspase-3 protein in 15 normal colonrectal mucosa and 66 colonrectal carcinoma specimens.Results Expressions of Bag-1 in normal colorectal mucosa tissues and colorectal cancers were 13.3%、74.2% respectively.Expressions of Caspase-3 in normal colorectal mucosa tissues and colorectal cancers were 46.7%、68.2% respectively.In colorectal carcinoma,Bag-1 expression was related to differentiation degree,lymph node metastasis and Duke's stage(P0.05).Caspase-3 expression was not related to differentiation degree,lymph node metastasis,and Duke's stage(P0.05).The expression of Bag-1 in colorectal carcinoma had obvious correlation with Caspase-3 expression(P0.05).Conclusions Bag-1 and Caspase-3 may play an important role in development of conlonrectal carcinoma and Bag-1 can be used as new indicators for judging biological behaviour of colontectal carcinoma.
