摘要 研究克隆了秦艽3-羟基-3-甲基戊二酰辅酶A还原酶(HMGR)家族成员的两条基因,并对其序列进行了生物信息学分析.结果显示,两条HMGR基因都包含完整的开放读码(ORF)框,编码蛋白含有HMGR蛋白特有的4个保守的活性位点,与其他植物HMGR蛋白序列具有较高的一致性,都具有跨膜结构域,高级结构类同;GmHMGR1和GmHMGR2分别定位于细胞质膜和线粒体中.表达分析显示,GmHMGR1主要在根和花中表达,而GmHMGR2主要在叶中表达. Gentiopicroside, a secoiridoid glycoside, is one of the main pharmacological compounds of traditional Chinese medicine Gentiana. In this study, two Gentiana macrophylla HMGR genes were cloned based on our laboratory's high-throughput sequencing, and bioinformatics analysis was carried out. The results showed that two HMGR genes both contain the completed ORF, and their encoded proteins with two transmembrane domains and four HMGR protein-specific conserved active sites, high consistency with the other plant in HMGR protein sequences and high structure, but GrnHMGR1 and GmHMGR2 locate respectively in the cell plasma membrane and mitochondria, Quantitative PCR analysis showed that GmHMGRI has high expression levels in the roots and flowers in G. macrophylla, while GmHMGR2 mainly expresses in leaves.
