摘要
目的 :观察风疹病毒 ( RV)分离株在人的神经细胞中形态和形态发生过程。方法 :利用超薄切片电子显微镜技术对自行分离的 RV JR2 3 株在人脑神经细胞中的形态与形态发生过程进行了研究。 结果 :RV JR2 3 株感染人脑神经细胞 2 4 h后 ,可在细胞浆的核周围观察到病毒相关结构 ,包括电子致密颗粒 ,1 0 nm,核衣壳样结构 ,1 5~ 2 5nm,“包涵体”样结构 ,“条纹”样结构伴随核衣壳样颗粒 ,并逐渐增多。 96h可见完整病毒颗粒 ,1 92 h达到高峰。病毒颗粒呈圆形 ,由单层脂质膜包绕 ,直径 60~ 80 nm。病毒在双层核膜中间装配 ,然后披上单层核膜而成熟 ,核膜破碎后释放入细胞浆。病毒颗粒多见于核周围。结论 :RV JR2 3 株能在人神经细胞中广泛增殖 ,其形态发生有独特的方式。
Objective: To investigate morphology and morphogenesis of the isolated rubella virus(RV) in human cerebral neural cells.Methods: Ultrathin section electron microscopy was used to study replication, morphology and morphogenesis of RV JR 23 in human cerebral neural cells.Results: The virus related structures, including electron dense cores, 10 nm, nucleocapsid like structures, 15 25nm “inclusion body” like and “stick” like structures, were observed in the cytoplasm near the nucleus at 24h postinfection and gradually increased. Intact virions were observed at 96h and the peak appeared at 192h postinfection. The virion was spherical, 60 80nm, with a mono layered dense envelope. Virions were assembeled between the two layers of the nucleus membrane and released into the cytoplasm by breaking the membrane and enveloped with monolayered membrane. Therefore, the virions were usually seen around the nucleus.Conclusion: RV JR 23 can reproduce massively in the human cerebral neural cells and has its own distinctive morphogenesis.
