重组胸腺素α1对流感疫苗的增效作用 被引量:2

摘要 目的探讨重组胸腺素α1(Recombinant human thymosinα1,rhTα1)对免疫低下小鼠接种流感疫苗后免疫应答效果的影响。方法将BALB/c小鼠随机分为5组:疫苗组、环磷酰胺+疫苗组、环磷酰胺+疫苗+rhTα1低浓度(0.2 mg/kg)组、环磷酰胺+疫苗+rhTα1中浓度(0.4 mg/kg)组、环磷酰胺+疫苗+rhTα1高浓度(0.8 mg/kg)组。经小鼠腹腔注射环磷酰胺,80 mg/kg,共3次,隔天给药;肌肉接种流感疫苗,1.8μg血凝素/只;皮下注射低、中、高浓度rhTα1,0.2 ml/只,每周2次,连续4周。动态监测各组小鼠血清血凝抑制(HI)抗体效价,并观察小鼠脾脏指数及脾脏形态学结构的变化。结果与疫苗组相比,环磷酰胺+疫苗组小鼠体内HI抗体效价降低,脾指数降低,脾脏组织病理形态退化;与环磷酰胺+疫苗组相比,环磷酰胺+疫苗+rh-Tα1各浓度组HI抗体效价升高,小鼠脾指数增加,脾脏组织病理形态明显改善。结论重组胸腺素α1可提高免疫低下小鼠流感疫苗接种的免疫效果,为寻找适合免疫低下人群的疫苗增强剂提供了有益的尝试。 Objective To investigate the effect of recombinant human thymosin α1(rhTα1) on immune response induced by influenza vaccine in immunocompromised mice.Methods BALB / c mice were randomly divided into five groups,i.e.vaccine,cyclophosphamide + vaccine,cyclophosphamide + vaccine + rhTα1 at low dosage(0.2 mg / kg),cyclophosphamide + vaccine + rhTα1 at moderate dosage(0.4 mg / kg) and cyclophosphamide + vaccine + rhTα1 at high dosage(0.8 mg / kg) groups,8 for each.Mice were injected i.p.with cyclophosphamide at a dosage of 80 mg / kg every other day for 3 times.Influenza vaccine was injected i.m.at a dosage of 1.8 μg of hemagglutininper mouse.The rhTα1 at low,moderate and high dosages were injected s.c.in a volume of 0.2 ml,twice a week for 4 weeks.The HI antibody titers in sera of mice in various grousp were monitored dynamically,while the spleen index and morphology were observed.Results Compared with those in vaccine group,the serum HI antibody titer and spleen index of mice in cyclophosphamide + vaccine group decreased,while the morphological degeneration of spleen was observed.However,compared with those in cyclophosphamide + vaccine group,the serum HI antibody titers and spleen indexes in cyclophosphamide + vaccine + various dosages of rhTα1 groups increased,while the morphology of spleen were improved significantly.Conclusion The rhTα1 enhanced the immune effect of influenza vaccine in immunocompromised mice,which provided a useful attempts in search of vaccine enhancer suitable for immunocompromised populations.
