摘要
目的:通过观察苏木乙酸乙酯提取物对大鼠同种异位心脏移植慢性排斥反应血清中IL-10水平的影响,为进一步探讨苏木乙酸乙酯提取物对慢性排斥反应的作用机制奠定工作基础。方法:以Wistar大鼠为供体,以SD大鼠为受体,建立大鼠腹腔异位心脏移植模型。将手术成功后连续灌服21天环孢素A的大鼠,随机分为对照组、苏木组、环孢素A组,对照组按体质量计算等容量橄榄油灌胃;环孢素A组按12 mg/kg/d环孢素A溶于橄榄油给药;苏木组按含生药量1.15g/kg/d溶于橄榄油中给药。灌胃7天后采血,用ELISA法检测血清中IL-10的含量。结果表明:1)与对照组相比,环孢素A组与苏木组血清中IL-10的含量明显增高(P<0.01,P<0.05)。2)环孢素A组与苏木组相比有显著差异(P<0.01)。结论:苏木乙酸乙酯提取物能够升高移植心脏的大鼠血清IL-10的含量,这可能是抗慢性排斥反应的原因之一。
Objective:To observe the effect of ethyl acetate extracted from the sappan woou on rat neterotopic carmac transplantation for chronic rejection in serum IL - 10 levels, for the further study of sappan wood ethyl acetate extract on chronic rejection mechanism laying foundation. Methods:The animal model of abdominal cardiac ectopic transplantation was established by taking the Wistar rat as the donor and the SD rat as the receptor. Establishment of rat heterotopic heart transplantation model. After the success of the operation of continuous gavage for 21 days of CsA in rats, they were randomly divided into the model group,the SWE group and the CsA group. ELISA method was used to detect the content of IL - 10 in serum. Results : 1 ) Compared with the control group, serums IL - 10 of CsA group and SWE group were sig- nificantly increased ( P 〈 0.01, P 〈 0.05 ). 2 ) There was a significant difference between CsA group and SWE group (P 〈 0. 01 ). Conclusion:The ethyl acetate extract from sappan wood can improve cardiac allograft in rat serum IL - 10 content, which may be one of the reasons against chronic rejection.
