摘要
目的研究白介素-10(IL-10)基因G1082A和C819T单核苷酸多态性(SNP)与移植术后外周血微量巨细胞病毒(CMV)DNA的关系。方法采集176例移植术后受者,429例次的外周血。通过巢式聚合酶链反应(nPCR)技术检测微量CMV DNA。通过序列特异性聚合酶链反应(SSP-PCR)技术检测IL-10基因G1082A与C819T的变异情况。分析IL-10各基因型在微量CMV DNA受者中的分布频率,并对阳性组与阴性组进行比较。结果研究人群中IL-10G1082A和C819T基因型分布符合Hardy-Weinberg遗传平衡定律。CMV DNA阳性组IL-10G1082A和C819T等位基因频率分别为G=3.75%、A=96.25%和C=32.50%、T=67.50%;CMV DNA阴性组分别为G=5.51%、A=94.49%和C=36.03%、T=63.97%。CMV DNA阳性组两SNP基因型及等位基因分布与其在CMV DNA阴性组中的分布差异无显著性(P>0.05)。结论目前尚不能认为IL-10基因G1082A与C819T单核苷酸多态性与移植受者微量CMVDNA阳性有相关性。
Objective To investigate the relationship between the IL-10 G1082A and C819T 's SNP and the trace CMV DNA of recipients.Methods The trace CMV DNA in 429 samples of 176 patients was detected by nested polymerase chain reaction (nPCR). The IL-10 G1082A and C819T's SNP were determined using sequence specific primers polymerase chain reaction (SSP - PCR). Results In the 40 CMV DNA - positive samples, the G1082A allele frequencies were 3.75 % for G mid 96.25 % for A, with the genotype frequencies 0 % for G/G, 92.5 % ( n = 37 ) for A/A, and 7.5 % (n=3) for G/A; In the 136 CMV DNA- negative samples, the G1082A allele frequencies were 5.51% for G and 94.49% for A, with the genotype frequencies 0% for G/G, 88.97% ( n = 121) for A/A, and 11.03% ( n = 15) for G/A. In the CMV DNA - positive group, the C819T allele frequencies were 32.5% for C and 67.5% for T, with the genotype frequencies 5% ( n = 2) for C/C, 40% ( n = 16) for T/T, and 55% ( n = 22) for C/T; In the CMV DNA - negative group, the IL - 10 C819T allele frequencies were 36.03% for C and 63.97% for T, with the genotype frequencies 11.76% ( n = 16) for C/C, 39.71% ( n = 54) for T/T, and 48.53 % ( n = 66) for C/T. The two SNPs distribution of the frequencies were all statistically similar between the CMV DNA - positive/negative groups( P 〉 0.05). Conclusions The IL- 10 gene G1082A and C819T's SNP are not associated with the CMV DNA - positive recipients.
