转HJC-1和G6-EPSPS基因抗虫耐除草剂大米对BN大鼠致敏性评价

Assessment of allergenicity of rice genetically modified with HJC-1 and G6-EPSPS genes by BN rats model

摘要 目的利用BN大鼠对转HJC-1和G6-EPSPS基因抗虫耐草甘膦大米进行致敏性评价。方法 42只BN大鼠随机分为7组,每组6只,分别为溶剂对照组、饲养对照组、亲本大米组、转基因大米组、HJC表达蛋白组、G6表达蛋白组和阳性对照组。转基因大米组喂饲含70%转HJC-1和G6-EPSPS基因大米饲料,亲本大米组喂饲含70%的9746亲本大米饲料,表达蛋白组每天每只动物分别灌胃给予1.0 mg/ml HJC和G6蛋白各1 ml,阳性对照组每天每只灌胃给予1.0 mg/ml OVA溶液1 ml,溶剂对照组每天每只灌胃给予PBS 1 ml,连续42 d。各组动物饲料均不含卵蛋白。观察动物体重、行为、毛色、分泌物等毒性表现及死亡情况,每周称重并记录体重,分别于第28天和第42天使用ELISA试剂盒测定血清抗体IgG和IgE水平,试验结束时称重并计算脾和胸腺脏器系数。结果实验期间各组动物进食正常,活动自如,未出现生病或死亡,各组动物体重及总增重差异无统计学意义。与溶剂对照组和饲养对照组比较,HJC和G6表达蛋白组、转基因大米组血清IgG、IgE抗体水平及脾和胸腺脏器系数差异均无统计学意义。阳性对照组血清IgG、IgE抗体水平高于溶剂对照组,差异有统计学意义(P<0.05)。结论全食品喂饲及灌胃给予表达蛋白未发现转HJC-1和G6-EPSPS基因抗虫耐草甘膦大米对BN大鼠血清IgG和IgE抗体水平及免疫器官产生不良影响,未见其对BN大鼠具有潜在致敏性。 Objective To assess the allergenicity of rice genetically modified with HJC-1 and G6-EPSPS genes by BN rats. Methods A total of 42 female BN rats were randomly divided into seven groups (six for each group), solution control group, breed control group, parent rice group, genetically modified rice group (GM group), HJC group, G6 group and positive control group (OVA). GM group and parent rice group were fed with diets containing rice genetically modified with HJC-1 and G6-EPSPS genes and 9746 rice with mass fraction of 70% respectively for 42 days. HJC group, G6 group and positive control group were daily administrated 1 ml per rat at the concentration of 1 mg/ml HJC-1, G6-EPSPS and OVA by gavage, while the solution control group were given 1 ml PBS per rat. All of them were fed with diets without eggs. The general behavior, toxic performance and body weight were observed. The serum was collected on the 28 th day and the 42nd day for determination of IgG and IgE. In addition, immune organs were taken for calculating their organ coefficients. Results During the experiment, the BN rats exhibited free movements without death or sickness. There were no adverse effects on body weight or immune organ coefficients in all of the groups. Compared with negative group, there were no significant differences(P〉0.05 ) in the levels of serume IgG and IgE in GM group, HJC group and G6 group. The levels of serum IgG and IgE in positive group were higher than those in negative group (P〈0.05). Conclusion No IgG or IgE responses were elicited by rice genetically modified with HJC-1 and G6-EPSPS genes, while OVA elicited strong IgG and IgE responses. No allergenicity of rice genetically modified with HJC-1 and G6-EPSPS genes was found in BN rats model.
