摘要
目的:检测结直肠癌患者外周血循环中黑素瘤抗原编码基因(melanoma antigen encoding gene,MAGE)-A3、MAGE-A6和MAGE-A10的表达情况,并分析其临床意义。方法:采用实时荧光定量PCR(real-time fluorescent quantitative PCR,RFQ-PCR)检测35例结直肠癌患者和15例正常对照者外周血中MAGE-A3、MAGE-A6和MAGE-A10mRNA的表达水平,并分析其与结直肠癌患者临床病理特征(性别、年龄、肿瘤部位、组织学分级、肿瘤浸润深度、淋巴结转移和Dukes分期)之间的关系。结果:所有外周血样本中均未检测出MAGE-A3mRNA的表达。结直肠癌组MAGE-A6mRNA表达阳性率显著高于正常对照组(P=0.0000),男性患者的表达水平高于女性患者(P=0.0459),淋巴结转移患者的表达水平也高于无转移患者(P=0.0000);结直肠癌组MAGE-A10mRNA表达阳性率也显著高于正常对照组(P=0.0224);淋巴结转移患者的表达水平也高于无转移患者(P=0.0000)。结论:结直肠癌患者外周血循环中MAGE-A6和MAGE-A10mRNA的表达在结直肠癌转移检测中具有重要意义,可能成为结直肠癌微转移诊断、治疗选择和预后判断中的重要临床靶标。
Objective: To analyze the mRNA expression levels of MAGE (melanoma antigen encoding gene)-A3, MAGE-A6 and MAGE-A10 in peripheral blood of patients with CRC (colorectal carcinoma), and to study their clinical significance. Methods: Peripheral blood samples were collected from 35 CRC patients and 15 healthy donors. RFQ-PCR (real-time fluorescent quantitative PCR) was performed to detect the mRNA expression levels of MAGE-A3, MAGE-A6 and MAGE-A10 for all samples. The correlation of clinicopathological characteristics (gender, age, primary tumor lesion, histologic grading, depth of invasion, lymph node metastasis, Dukes’ staging) with the expressions of MAGE-A6 and MAGE-A10 mRNAs in peripheral blood of patients with CRC (colorectal cancer) was evaluated. Results: MAGE-A3 mRNA expression was not detected in all peripheral blood samples. The positive rate of MAGE-A6 mRNA in CRC group was significantly higher than that in healthy control group (P = 0.000 0), and its expression level in CRC male patients was significantly higher than that in female patients (P = 0.045 9). Furthermore, MAGE-A6 mRNA was more significantly expressed in CRC patients with lymph node metastasis compared with the patients without lymph node metastasis (P = 0.000 0). The positive rate of MAGE-A10 mRNA in CRC group was also significantly higher than that in healthy control group (P = 0.022 4), and the expression level in CRC patients with lymph node metastasis was significantly higher than that in patients without lymph node metastasis (P = 0.000 0). Conclusion: The mRNA expression level of MAGE-A6 and MAGE-A10 in peripheral blood of CRC patients shows an important significance for CRC metastasis detection, and it may become a potential clinical marker for the diagnosis of micro-metastasis in lymph nodes, treatment selection and prognostic judgement of CRC.
