摘要
目的通过检测体外授精-胚胎移植(IVF-ET)受孕与自然受孕胎盘组织中胰岛素样生长因子-Ⅰ、Ⅱ(IGF-Ⅰ、Ⅱ)的表达,了解IVF-ET受孕对IGF-Ⅰ、Ⅱ表达的影响。方法选择2007年1月至2009年1月,不孕因素均为输卵管因素,辅助生育技术方法为IVF-ET的49例产妇为研究组,选择同期49例健康的自然受孕孕妇为对照组。采用RT-PCR技术检测两组胎盘组织中IGF-ⅠmR-NA、IGF-ⅡmRNA表达水平,免疫组化技术检测两者胎盘组织中IGF-Ⅰ、IGF-Ⅱ蛋白含量。结果研究组胎盘中IGF-Ⅰ mRNA、IGF-ⅡmRNA相对表达量依次为0.30±0.13、0.28±0.04;对照组胎盘中IGF-ⅠmRNA、IGF-ⅡmRNA表达量为0.65±0.10、0.91±0.26,研究组IGF-Ⅰ、IGF-Ⅱ平均光密度为0.26±0.04、0.29±0.05;对照组IGF-Ⅰ、IGF-Ⅱ平均光密度为0.32±0.07、0.34±0.04。研究组胎盘中IGF-ⅠmRNA、IGF-ⅡmRNA和蛋白表达水平均低于对照组,差异有统计学意义(P〈0.05)。研究组低出生体重儿发生率8.16%,对照组低出生体重儿发生率2.04%,差异无统计学意义(P〉0.05)。结论体外授精一胚胎移植孕妇胎盘中IGF-Ⅰ及IGF-Ⅱ表达水平低于自然受孕孕妇,但两组产妇新生儿出生体重相近,且低出生体重儿比例相当,提示IGF-Ⅰ、Ⅱ有促进胎儿的生长发育的作用,对胎儿的生长发育影响不大。
Objective To study the expression changes of placental insulin-like growth factor I and Ⅱ (IGF-Ⅰ and IGF-Ⅱ) between pregnancies following the in vitro fertilization and embryo transfer ( IVF - ET) and those conceived spontaneously. Methods The placenta were collected from 49 cases of pregnancies after IVF - ET( case group) and 49 cases of pregnancies who were normal (control group). The expressions of IGF-Ⅰ and IGF-1I mRNA in placenta were detected by reverse transcription- polymeras chain reaction (RT-PCR). The levels of IGF-Ⅰ and IGF-Ⅱ protein were determined by immunohistochemistry. Results The mRNA levels of the placental IGF-Ⅰ and IGF Ⅱ were 0. 30 ±0. 13 and 0. 28 _+0. 04 in the IVF-ET group, and 0. 65 ±0. 10 and 0. 91 +0. 26 in the control group. The protein levels of the placental IGF-Ⅰ and IGF-Ⅱ were 0. 26 ±0. 04 and O. 29 ± 0. 05 in the IVF-ET group, and O. 32 ± 0. 07 and O. 34 ± 0. 04 in the control group. The mRNA and protein expressions of IGF-Ⅰ and IGF-Ⅱ in human placenta were significantly decreased in IVF-ET group compared to control group ( P 〈 0. 05 ). The incidence rate of low birth weight infant in IVF-ET group was significantly higher than control( P 〉0. 05). Conclusions Expressions of placental 1GF-Ⅰ and IGF-Ⅱ in the IVF-ET group did not affect fetal growth and development.
