摘要
目的:建立同时测定苦地丁中6种生物碱成分的HPLC含量测定方法。方法:采用高效液相色谱法,KromasilC18色谱柱(4.6 mm×250 mm,5μm),以0.02 mol.L-1磷酸二氢钾水溶液-甲醇为流动相梯度洗脱,流速0.8 mL.min-1,检测波长289 nm,柱温为室温。结果:6种生物碱在一定范围内线性关系良好(r>0.999),平均回收率为98.26%~102.10%,RSD均<3.0%。结论:该方法可以同时测定苦地丁中6中生物碱的含量,为苦地丁的质量评价提供参考。
Objective: To establish a HPLC method for the simultaneous determination of six alkaloids ( dehydrocheilanthifoline, protopine, corynoline, acetylcorynoline, oxysanguinarine, dihydrosanguinarine ) in Corydalis bungeana. Method: The chromatographic separation was performed on a Kromasil Cls column (4.6 mm ~250 mm, 5 ~xm) at room temperature with the 0.02 mol ·L^(-1) KH2PO4-methanol as the mobile phrase in gradient elution. The detection wavelength was set at 289 nm and the flow rate was 0. 8 mL ·min^(-1). Result: All the compounds were linear (r 〉 0. 999) in the range of the test concentrations, and the average recovery of the method was between 98.26% and 102. 10% , RSD 〈 3.0%. Conclusion: The method established by this paper can be used to determine six alkaloid components in C. bungeana simultaneously, and provides a scientific basis for the quality evaluation of C. bungeana.
