摘要
利用甘露糖-Sepharose 4B亲和柱从半夏总蛋白中分离纯化得到一种12kD的半夏凝集素,经质谱分析明确其为半夏凝集素单体。将该12kD凝集素单体作用于体外培养的HeLa细胞,MTT法检测细胞活性并用倒置相差显微镜观察凝集素对HeLa细胞生长状态的影响。结果显示:低浓度(0.004、0.02、0.1mg/mL)半夏凝集素具有一定促进HeLa细胞增殖的作用;高浓度(0.5、1mg/mL)半夏凝集素有一定抑制Hela细胞增殖的作用。
One kind of Pinellia ternata lectin is isolated from the total protein of Pinellia ternata via mannose-Sepharose 4B affinity chromatography. The cultured Hela cell added with different concentrations of lectin is detected via the MTT method and observed with an inverted phase contrast microscope. The re- sults showed that low concentrations (0. 004 mg/mL, 0.02 mg/mL, 0. 1 mg/mL) of Pinellia ternata lec- tin can promote the proliferation of Hela cells, but high concentrations (0. 5 mg/mL, 1 mg/mL) of Pinellia ternata lectin can inhibit the proliferation of Hela cells.
