摘要
目的建立测定蒲公英注射液中咖啡酸含量的高效液相色谱法。方法使用XTerra MS C18(4.6 mm×150 mm,5μm)色谱柱,以甲醇-乙腈-磷酸盐缓冲液(18∶2∶80,V/V)为流动相,流速1.0 ml.min-1,柱温25℃;在323 nm检测波长条件下,考察方法的专属性、准确度、精密度、稳定性等。结果咖啡酸进样量在0.02~0.50μg范围内与峰面积呈现良好的线性关系,回归方程为Y=1 695.52 X+6.158 2×106(r2=0.999 9);平均加样回收率为99.04%,RSD=0.75%。结论该法简单、准确、稳定,可以用于蒲公英注射液中咖啡酸的含量测定。
Objective To establish an HPLC method for the determination of caffeine acid in taraxacum injection. Methods The HPLC method was validated using a XTerra MS Cls analytical column (4.6 mmx 150 mm ,5μm) and the mobile phase of methanol acetoni trile phosphate buffer( 18:2: 80, V/V) at a flow rate of 1.0 ml·min- 1 with UV detection at 323 nm. The column temperature was kept at 25℃. The validation parameters including precision, repeatability, stability etc were observed. Results The linear range of caffeic acid was from 0.02 to 0.50 μg. regression equation was Y = 1 695.52X + 6. 158 2 x 10s ( r2 = O. 999 9 ). The average recovery was 99.04%, and RSD was 0.75%. Conclusion This method is simple, convenient, accurate, and was good stability with good repeat- ability, which can be used for the determineation of caffeine acid in taraxacum injection.
