摘要 目的:探讨核因子-κB(NF-κB)特异性抑制剂吡咯烷二硫代氨基甲酸盐(PDTC)对鼠黑素瘤B16细胞增殖及血管内皮生长因子(VEGF)表达的影响。方法:不同浓度的PDTC作用于B16细胞不同时间后,MTT法测定其对B16细胞增殖的抑制率,ELISA检测PDTC作用下B16细胞VEGF的表达情况。结果:经PDTC处理的实验组,肿瘤细胞增殖活性明显受到抑制(P<0.01);与对照组相比,PDTC作用后B16细胞VEGF表达降低(P<0.01)。结论:PDTC具有显著抑制鼠黑素瘤B16细胞生长及VEGF表达的作用,是一种有潜力的抑制黑素瘤增殖的药物。 Objective:To explore the effects of pyrrolidine dithiocarbamate(PDTC)on the proliferation of mouse melanoma cell line B16 and expression of vascular endothelial growth factor(VEGF). Methods:B16 cells were treated with PDTC by different concentration and different time in vitro.The inhibition rates of cell growth were assayed by MTT method.The expression of VEGF was detected by ELISA. Results:The tumor cells proliferation activity of the experimental group with PDTC processing were inhibited obviously(P〈0.01) in a time and dose dependent.Compared with the control group,the expression of VEGF of PDTC group cells was downregulated in a dose-dependent manner(P〈0.01). Conclusions:PDTC can inhibit the growth and VEGF expression of B16 cells,it may be a potential chemotherapeutics for the proliferation of melanoma.
