摘要 [目的]观察核因子-κB(NF-κB)抑制剂吡咯烷二硫代氨基甲酸盐(PDTC)对小鼠肝癌Hca-F细胞生长的影响。[方法]用含PDTC的完全培养基体外培养Hca-F细胞,用MTT法测Hca-F细胞增殖抑制率。体内实验中所有小鼠均接种Hca-F并随机分为两组:攻击对照组腹腔注射生理盐水,PDTC治疗组腹腔注射PDTC溶液,每天测量肿瘤体积和体重。治疗后第10天处死小鼠,取肿瘤组织。用MDA试剂盒测定瘤组织MDA含量,用放射免疫法测定肿瘤组织TNF-α的含量。[结果]PDTC在体外可抑制Hca-F增殖,浓度为25μg/mL时细胞增殖抑制率为85.4%。体内实验中PDTC治疗组小鼠肿瘤体积及重量、瘤组织匀浆MDA和TNF-α含量,与对照组比较差异均无显著性意义(P>0.05)。[结论]PDTC在体外对肝癌细胞增殖有抑制效应。 [Objective] To observe the effects of pyrro1idine dithiocarbamate(PDTC),an inhibitor of NF—κB,on the growth of mouse Hca-F cell in vitro and in vivo.[Methods] Hca-F cells were cultured in complete medium containing PDTC in vitro.MTT assay was used to detect the inhibition of growth of Hca-F cell.In vivo,the mice were inoculated with Hca-F cells and divided randomly into two groups: mice of challenge control group were injected with normal saline,while those of PDTC treatment group were injected with PDTC solution.The volume of tumor and body weight of mice were measured every day.Mice were killed on day 10 after treatment.Malondialchehyche(MDA) content of tumor tissue was assayed with MDA reagent kit and TNF-content was assayed with RIA.[Results] PDTC inhibited the proliferation of Hca-F cells in vitro,the proliferation inhibition rate was 85.4% on concentration 25 μg/mL.In vivo,differences in the volume,weight of tumor,content of MDA and TNF-α level of tumor tissue supernant between the two groups were not significant(P〉0.05).[Conclusion] PDTC can inhibit the proliferation of Hca-F cell in vitro.
