摘要 目的观察骨髓间充质干细胞在无诱导因素干预下,自身表达平滑肌肌动蛋白(smooth muscle alpha-actin,α-actinSM)情况。方法采用直接贴壁法培养SD大鼠骨髓间充质干细胞,传代培养至第4代时,对间充质干细胞进行鉴定。利用抗α-actin SM荧光抗体,检测间充质干细胞胞质内α-actin SM表达,计算其阳性率。结果骨髓间充质干细胞在无诱导因素干预下,其自身即可表达平滑肌早期特异性标志α-actin SM,阳性率高达44.1%。结论骨髓间充质干细胞体外培养时,无需诱导因素干预,能够自发向平滑肌细胞方向分化。 Objective To observe bone marrow mesenchymal stem cells cultured in absence of inducing factors self-express smooth muscle alpha-actin (a-actin SM). Methods Using adherent method to culture SD rat bone marrow mesenchymal stem cells in vitro culture to the fourth generation, then the mesenchymal stem cells were examined, a-actin SM expression of MSCs was analyzed with fluorescence immunocytochemistry by using anti-a-actin SM antibody. Results Without inducing factors, bone marrow mesenchymal stem cells could self- express smooth muscle's early specific markers of a -actin 44.1%. Conclusion Bone marrow mesenchymal stem cells differentiate into smooth muscle cells. SM, the positive rate of a-actin SM expression was cultured without inducing factors can spontaneously
