摘要
目的 :从森林及常年堆放木材的地方采集样本 ,分离出有较强分解纤维素能力的木霉 ,诱变后使酶活性提高。方法 :用选择性培养基进行分离培养 ,常规方法鉴定 ,用HNO2 和UV进行联合诱变。结果 :3 5份样品中分离出木霉12株 ,其中一株绿色木霉 (TrichodermaViride)有较强分解纤维素的能力 ,诱变处理后CMc酶活性提高 90 % ,FP酶活性提高 75 % ,粗酶液 10 0 %崩解滤纸时间由原来的 14小时缩短为 4小时 ,结论
objective:To select and culture cellulase productive Trichoderma from samples and to acquire a cellulase overproducing strain by mutagenesis.Methods:The samples were isolated by selective culture medium ,and the fungi were identified by the routine method.Then the isolated strains were mutated with HNO 2 and UV;Optimum rection conditions for the activity of the enzyme were investigated.Meanwhile,the enzymogical property were tested.Results:12 strains of Trichoderma were isolated from 35 samples.One of the 12 strains had stronger ability to break down cellulose.It was identified as Trichoderma Vivide.The activites of CMCase and FPase increased 90% and 75% after mutagenesis.Conclusion:The isoleted strain has a applied value.
