摘要
目的:观察缬草提取物(VOL)预处理对大鼠离体心脏缺血再灌注(I/R)损伤和相关生化指标以及细胞内游离钙的影响,探讨VOL抗I/R损伤的机制。方法:利用Langendorff建立大鼠离体心脏I/R模型,观察左心室发展压(LVDP)恢复和再灌注痉挛度。用生化技术检测冠脉流出液中乳酸脱氢酶(LDH)、磷酸肌酸激酶(CK)的活性以及心肌细胞线粒体超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSHPx)、ATP酶(ATPase)活性和丙二醛(MDA)含量,并用激光扫描共聚焦显微镜,借用Fluo-3/AM荧光染色技术观察心肌细胞内Ca2+强度。结果:①VOL预处理对心肌I/R损伤具有明显保护作用,并随浓度增加,对心脏保护作用逐渐增强,再灌注痉挛度明显减弱,心脏从缺血后恢复跳动更迅速,心律失常的发生越来越少,与I/R组相比,100mg/L VOL使LVDP的恢复从(20.35±2.81)%增加到(91.87±6.52)%(P<0.01),再灌注痉挛度从(89.27±9.31)mmHg减轻为(12.23±3.51)mmHg(P<0.01)。②VOL预处理明显降低LDH、CK活性和MDA含量,显著增加SOD、GSHPx、ATPase的活性,且随VOL浓度增高,上述作用更加明显,100mg/L VOL预处理组各项生化指标与对照组无明显差异。③VOL使I/R损伤后心室肌细胞内Ca2+荧光强度明显减少,随着VOL浓度增高,减少细胞内Ca2+的作用更加强烈。与I/R组相比,100mg/L的VOL使心肌细胞内Ca2+荧光强度从144.85±16.41减少到了51.83±11.61(P<0.01)。结论:VOL预处理能明显抵抗I/R损伤,可能机制与VOL减少心脏I/R时心肌细胞内游离钙浓度的增加和抗脂质过氧化有密切关系。
Objective: To investigate the protecting effect of valeriana officinalis L extract(VOL)-preconditioning on ischemia-reperfusion(I/R) injury,and to explore the impact on the related biochemical factors and free Ca^2+ increase of cardiomyocytes in isolated rat hearts.Methods: The hearts models of I/R in rats in vitro were set up through using Langendorff-perfusion system,and the recovery of left ventricular developed pressure(LVDP) and magnitude of reperfusion contracture from I/R were observed.The levels of lactate dehydrogenase(LDH) and creatine kinase(CK) in coronary flowing solution,and the activities of superoxide dismutase(SOD),malondialdehyde(MDA),adenosine triphosphate(ATP)-ase,glutathione peroxidase(GSHPx) from cardiomyocyte mitochondria,were determined by biochemical technology.The fluorescence intensity of intracellular Ca^2+ in cardiomyocytes of rats was detected with Flup-3/AM loading by the laser scanning confocal microscopy.Results: ①VOL markedly antagonized I/R injury in a dose-dependent manner in isolated rat hearts.Compared with I/R group,VOL(100 mg/L) significantly recovered the LVDP from(20.35±2.81) % to(91.87±6.52) %(P〈0.01),and decreased the reperfusion contracture from(89.27±9.31) mmHg to(12.23±3.51) mmHg(P〈0.01).In the VOL groups,occurrence of arrhythmia almost could not be induced by I/R.②VOL significantly reduced LDH and CK activities and MDA levels,and enhanced the activities of the SOD,ATP-ase,GSHPx in cardiomyocytes mitochondria in a concentration-dependent manner.And as the concentration of VOL increased to 100 mg/L,these biochemical indicators had no significant difference from those of control group.③VOL reduced Ca^2+ overloading concentration-dependently.Compare with I/R group,100 mg/L VOL significantly decreased the fluorescence intensity of intracellular Ca^2+ in cardiomyocytes of rats from(144.85±16.41) to(51.83±11.61)(P〈0.01).Conclusion: VOL could obviously protect the I
