加味茵芍散对肝纤维化家兔TGF-β1、Smad2、Smad4 mRNA表达的影响 被引量:4

摘要 目的:从加味茵芍散对肝纤维化家兔转化生长因子-β1(TGF-β1)、丝/苏氨酸激酶2(Smad2)、丝/苏氨酸激酶4(Smad4)mRNA表达的影响探讨该方对肝纤维化的可能作用机制。方法:按0.10mL/kg的用量腹腔注射CCl4诱导复制家兔肝纤维化模型,50只家兔随机分成模型组,秋水仙碱组,加味茵芍散高、中、低剂量组5组,每组10只,通过肝脏病理取材确定肝纤维化病变,同时实时定量逆转录一聚合酶链反应法检测TGF-β1、Smad2 mRNA、Smad4 mRNA的表达。结果:与模型组相比,秋水仙碱组和加味茵芍散高、中、低剂量组家兔肝组织TGF-β1、Smad2 mRNA、Smad4 mRNA表达明显减弱(P<0.05);加味茵芍散中、高剂量组与秋水仙碱组相比TGF-β1、Smad2 mRNA、Smad4 mRNA的表达量显著降低(P<0.05)。结论:加味茵芍散能减轻纤维组织增生,可以下调TGF-β1、Smad2 mRNA、Smad4的表达。该方对CCl4诱导家兔肝纤维化模型具有一定保护作用、可以延缓肝纤维化的进展,其作用机制可能与调节TGF-β/Smad信号通路有关。 Objective: To explore the effect of modified Yin Shao Powder on expression of TGF- β 1, Smad2, Smad4 in hepatic fibrosis rabbits. Methods: According to the amount of injected 0.10mL/kg copy CC14-induced hepatic fibrosis model in rabbits, 50 rabbits were randomly divided into five groups as model group, colchicine group, modified Yin Shao Powder high, medium and low dose groups, 10 rabbits for each group. Determined the liver fibrosis pathology by liver drawn, while real- time quantitative reverse transcription-polymerase chain reaction was used to detecte the TGF- β 1, Smad2, Smad4 and other indicators. Results: Compared with model group, the expression of TGF- β 1 in colchicine group and modified Yin Shao Powder high, medium and low-dose groups were significantly decreased (P〈0.05); compared with the colchicine group, the expression of Smad2, Smad4 in modified Yin Shao Powder were significantly lower (P〈0.05). Conclusion: Modified Yin Shao Powder can reduce fibrosis, can reduce the expression of TGF-β 1, Smad2, Smad4. This prescription has a protective effect of the CCI4- induced hepatic fibrosis model in rabbits, delay the progress of hepatic fibrosis, and its mechanism may be related to regulation TGF- β/Smad signaling pathway.
