摘要
目的研究盐酸米诺环素对脂多糖(LPS)作用下人牙周膜成纤维细胞(HPDLF)的白细胞介素-6(IL-6)mRNA表达的影响。方法本研究于2011年10月至2012年2月在井冈山大学医学院重点实验室进行。培养HPDLF并分为以下5组:对照组(未加药)、高剂量组(0.01g/LLPS+0.2g/L盐酸米诺环素)、低剂量组(0.01g/LLPS+0.05g/L盐酸米诺环素)、中剂量组(0.01g/LLPS+0.1g/L盐酸米诺环素)、模型组(0.01g/LLPS),每组均复种3孔。实时荧光定量PCR检测各组HPDLF中的IL-6mRNA表达情况。结果对照组、高剂量组、低剂量组、中剂量组、模型组的IL-6mRNA相对表达量分别为:0.61±0.08、0.62±0.10、0.88±0.17、0.49±0.21、1.88±0.07。模型组IL-6mRNA的相对表达量高于对照组、高剂量组、低剂量组、中剂量组,且差异有统计学意义(P<0.01)。对照组及低、中、高剂量组的IL-6mRNA相对表达量两两比较,差异均无统计学意义(P>0.05)。结论盐酸米诺环素能够降低LPS作用下HPDLF中的IL-6mRNA的表达,这可能是盐酸米诺环素治疗牙周炎的作用机制之一。
Objective To study the effect of minocycline hydrochloride on interleukin 6 (IL-6) mRNA expression of human periodontal ligament fibroblasts (HPDLF) under the influence of lipopolysaccharide(LPS). Methods This experiment was carried out in Key Laboratory of Medicine Institute of Jinggangshan University from October 2011 to February 2012. Human periodontal ligament fibroblasts were cultivated and five groups were established:control group , high group (0.01 g/L LPS+0.2 g/L minocycline hydrochloride), low group (0.01 g/L LPS +0.05 g/L minocycline hydrochloride ) , medium group (0.01 g/L LPS + 0.1 g/L minocycline hydrochloride ) and model group (0.01 g/L LPS ). Real-time fluorescence-based quantitative PCR was used to determine the expression of IL-6 mRNA of HPDLF cells induced by LPS. Results The expression of IL-6 mRNA relative expression in control group, high group, low group, medium group and model group was respectively: 0.61± 0.08, 0.62 ±0.10, 0.88 ±0.17,0.49±0.21 and 1.88±0.07. IL-6 mRNA relative expression of model group was higher than that in control group, high group, low group and medium group, and the difference was statistically significant (P 〈 0.01 ). IL-6 mRNA relative expression in control group, high group, low group and medium group were compared with each other, and the differences were not statistically significant (P 〉 0.05). Conclusion Minocycline hydrochloride can reduce LPS intervention in human periodontal ligament fibroblastIL-6 mRNA expression, which may be the functioning mechanism of minocycline hydrochloride in treatment of periodontitis.
