摘要
目的:通过梯度洗脱的方法,建立了如意金黄散中小檗碱、姜黄素、大黄素含量测定的高效液相色谱法。方法:采用Kromasil C18色谱柱,以乙腈-9.6%醋酸为流动相梯度洗脱,柱温25℃,检测波长422nm,流速:1.0mL.min-1。结果:在此条件下姜黄素类物能够得到很好的分离,小檗碱、姜黄素、大黄素的线性范围分别为2.55~51μg.mL-1(r=0.9999),4.135~82.7μg.mL-1(r=0.9997),0.6225~12.45μg.mL-1(r=0.9997)。平均回收率(n=6)分别为99.02%、98.04%、98.58%。结论:该方法简便,快速易行,能够更好的控制如意金黄散的质量。
Objective:To establish a HPLC method for quantitative determination of berberine,curcumin and emodin in Ruyi Jinhuang power using gradient elution method.Methods:HPLC method was used,with Kromasil C18 column,acetonitrile-9.6%acetic acid,linear gradient elution,column temperature was maintained at 25℃,as mobile phase with a flow rate of 1.0mL·min-1,and detecting wavelength 422nm.Results:In this chromatographic conditons,the curcumins were completely separated.The calibration curves of berberine,curcumin and emodin were linear in the range of 2.55~51μg·mL-1(r=0.9999),4.135~82.7μg·mL-1(r=0.9997),0.6225~12.45μg·mL-1(r=0.9997),respectively;The average recoveries(n=6)were 99.02%,98.04%,98.58%,respectively.Conclusion:The method is simple,fast and easy,and controling the quality of Ruyi Jinhuang power effectively.
