大孔吸附树脂分离纯化红枣总黄酮的研究

摘要 目的筛选出分离纯化红枣总黄酮的最佳大孔树脂型号及工艺条件。方法以静态吸附和解吸率为指标考察5种大孔树脂,优选出分离纯化红枣总黄酮的树脂类型,并确定分离纯化红枣总黄酮的工艺参数。结果 D101型大孔吸附树脂对红枣总黄酮的最佳吸附条件为:上样液质量浓度为0.403mg/mL,上样液pH值为3.0,吸附流速为3.0BV/h,上样液体积为4倍树脂体积,洗脱剂用80%的乙醇,洗脱流速为2.0 BV/h,洗脱剂的用量为6倍的树脂体积,分离纯化后红枣总黄酮的含量能达到19.5%。结论该研究为红枣总黄酮的吸附及脱附提供了实验研究数据。 Objective To establish an optimum separation and purification process of total flavonoids in Zizyphus jujuba by macroporous resins.Methods 5 models of macroporous resins were selected for purification test,the technical conditions for purification of the total flavonoids in Zizyphus jujuba with macroporous resins were studied by using the absorption of static and dynamic state.Results Experimental results of the static and dynamic adsorption-desorption experiments show that D101 resin possesses higher absorption and desorption capacity.The best result was based on the follows: the concentration of sample solution was 0.403 mg/mL,and its pH was 3.0,adsorption flow rate was 3.0 BV/h,the sample solution was 4 times as the volume of resin,the eluant was 80% alcohol and 8 times as the volume of the resin,elution flow rate was 2.0 BV/h,the amount of eluent was 6 times as the resin volume,the content of total flavonoids in the purified extracts reached 19.5 % with D101 in the optimized conditions.Conclusion This study offered theory evidences for adsorption and desorption of total flavonoids.
