摘要
目的探讨一种适合于临床应用的HPV-DNA分型检测方法。方法利用HPV保守区引物PCR扩增和型特异性探针膜芯片杂交,对包括5种低危型(HPV6、11、42、43、44)和18种高危型(HPV16、18、31、33、35、39、45、51、52、53、56、58、59、66、68、73、83、MM4)HPV进行联合分型检测,并将分型结果测序验证。结果在281例尖锐湿疣组织中发现265例HPV阳性(94.31%),PCR阳性标本均成功分型,其中低危型224例(84.53%),高危型41例(15.47%);单一型223例(84.15%),混合型(多重感染)42例(15.85%)。结论基因芯片检测HPV-DNA灵敏度高,特异性好,可在检出HPV感染同时显示亚型,适用于临床进行HPV感染的诊断及研究。
Objective To explore a clinical method for detection of HPV-DNA genotype in condyloma acuminate(CA) with gene-chips.Methods The gene-chips technique was used to determine 23 types which include 5 low risk and 18 high risk of HPV genotype in tissues from 281 patients with CA.Results There were 265 positive cases in 281 patients with CA,the total positive rates was 94.31%.The positive rates of low risk HPV genotypes was 84.35% with 224 cases,and high risk HPV genotypes was 15.47% with 41 cases.The positive rate from single infection with HPV was 84.15% with 223 cases and the mixed infection with HPV was 15.85% with 42 cases.Conclusion Gene-chips method is high sensitivities and specificities method in detection and typing of HPV.It is suitable for clinical diagnostic and research of HPV.
