不同海拔地区胎盘缺氧诱导因子在胎儿生长受限和巨大儿中的表达及意义 被引量:4

Expression and Its Significance of HIF-1α in Placenta in Fetal Growth Restriction and Macrosomia at Different Geographic Altitude

摘要 目的:缺氧诱导因子(HIF-1)是胎盘主要的调节氧代谢平衡因子,对细胞增殖、细胞凋亡、肿瘤生长、免疫有一定作用。检测不同海拔地区缺氧诱导因子(HIF-1)在胎儿生长受限和巨大儿患者胎盘组织中的表达情况,探索西宁地区胎儿生长受限和巨大儿的发病因素和发生的机制。方法:随机选择2010年1月—2011年2月在不同海拔地区(青海西宁、陕西咸阳)足月分娩的足月小儿(体重<2 500g的新生儿)各50例产妇为宫内发育受限组,选择同期分娩体重≥4 000g新生儿各50例产妇为巨大儿组;采用免疫组化链霉菌抗生物素蛋白-过氧化酶连接法(SP法)检测巨大儿和胎儿生长受限患者胎盘组织中缺氧诱导因子的表达。结果:平原地区巨大儿胎盘中HIF-1α蛋白表达下降,在胎儿生长受限患者胎盘组织中HIF-1α阳性表达不明显;但在西宁地区巨大儿胎盘中HIF-1α蛋白表达较平原地区上升,在胎儿生长受限患者胎盘组织中HIF-1α蛋白表达明显上升,差异有统计学意义(P<0.05)。结论:胎儿生长受限患者胎盘组织中HIF-1α蛋白表达明显上升,可能是胎儿生长受限发病机理中的一个重要因素。 Objective: To deal with the expression of HIF - 1α in placenta, as well as the pathogenesis and mechanism in fetal growth restriction (FGR) and macrsomia at different geographic altitude. Methods:We collected randomly 50 placentas of FGR ( 〈2 500g weight) and 50 placentas of maerosomia ( 〉14 000g weight) in Xining (2 260m altitude), Qinghai, and Xianyang (600m -850m ahtitude), Shanxi, respectively between Jan. 2010 and Feb. 2011. The Xining sample served as test group, Xianyang sample as control group, meanwhile, the samples were divided into FGR group and maerosomia group from the both group respectively. Expression of HIF - 1 α was detected by immunohistochemistry of Streptomyees biotin protein - peroxidase connection method (SP method). Results:In the control group, expression of HIF - 1α in macrosomia group was decreased, but no change of HIF - 1α for FGR group. Expression of HIF - 1α in maerosomia group was increased as compared with control group, and expression of HIF - 1α in FGR group was significantly increased ( P 〈 O. 05). Conclusions: Significant increased expression of HIF - 1α in FGR at high altitude may be an important factor for pathogenesis of FGR.
